In this work, an underlap channel-embedded field-effect transistor (FET) is proposed for label-free biomolecule detection. The device was fabricated by standard CMOS process, except for the gate-to-source/drain (S/D) alignment offset. The fabricated underlap device has a channel length of $10\microm$, a channel width of $20\microm$, and various underlap lengths from 100nm to 1400nm. Specifically, silica binding protein fused with avian influenza surface antigen (SBP-AIa) and avian influenza antibody (anti-AI) were designed as a receptor molecule and a target material, respectively. After the immobilization of SBP-AIa, the drain current increased slightly in comparison to that in the initial state. The small increment of the drain current can be explained by the presence of an additional fringing field due to the immobilized SBP-AIa, of which the thickness is approximately 3nm on the underlap region. However, by binding of the anti-AI molecules with the SBP-AIa at the underlap region, the drain current decreased considerably in all various underlap devices. The abrupt drop of the drain current is attributed to the negative charges of the anti-AI, and the decrement of the drain current becomes larger as the underlap length increases. To conform that the dramatic current reduction and the threshold voltage changes are due to the specific binding of SBP-AIa and anti-AI on the underlap region, a set of control experiments were carried out. As a result, only the binding of SBP-AIa and anti-AI effectively modulate the characteristics of underlap device and device also shows high specificity for detecting anti-AI biomolecule. Moreover, the drain current was returned back to its initial value after recovery test to break the binding sites of the biomolecules.
With the merits of a simple fabrication process, complementary metal-oxide-semiconductor compatibility, and enhanced sensitivity, the underlap FET could be a promising candidate for a chip-based biosensor. Furthermore, it has the capability to sense other biomolecules, including DNA, cancer markers, antigens and antibodies.
본 석사 논문에서는 underlap 구조의 MOSFET을 이용하여 바이오 센서로서의 동작 여부를 살펴보았다. Underlap 구조를 만들기 위하여 게이트와 채널 부분에 offset을 주어 공정을 진행하였으며, 검출 바이오 물질로써 조류독감 항체를 선정하여 실험을 진행하였다.
Underlap 구조 MOSFET의 전류-전압 특성을 살펴 본 결과 기존 MOSFET과 비교하여 오른쪽으로 이동된 형태의 그래프를 볼 수 있었으며 그에 따라 더 높은 문턱 전압을 가지는 것을 확인하였다. 이러한 특징은 underlap의 길이가 커짐에 따라 그 이동 정도가 더욱 커졌으며, 높은 드레인 전압 조건에서는 드레인 주변에서 공핍 영역이 형성됨에 따라 이러한 현상이 감소하는 것을 확인 하였다.
Underlap MOSFET의 바이오 센서로서 동작 여부를 살펴보기 위하여 조류독감 항원과 oxide와 결합력이 좋은 단백질이 합쳐진 형태의 SBP-AIa와 조류독감 항체인 anti-AI로 바이오 실험을 진행하였다. SBP-AIa가 underlap 부분에 결합된 후 특정 게이트 전압에서 드레인 전류는 약간 증가하였으며, 이는 SBP-AIa에 의한 fringing 전계의 증가현상으로 설명된다. 한편 anti-AI가 결합된 후에는 드레인 전류가 급격히 감소하였는데 이는 anti-AI가 가지고 있는 음성의 전하에 의해 underlap 부분의 inversion 현상이 더욱 힘들어 졌기 때문이다. 바이오 물질을 다시 떼어내는 recovery test, SBP-AIa와 특이 결합을 하지 않는 물질을 이용한 selectivity test, anti-AI와 같이 음성 전하를 띠는 DNA를 이용한 바이오 실험 결과를 통해 앞에서의 결과가 신뢰성이 있음을 증명하였다.
마지막으로 민감도를 높이기 위하여 underlap 부분에 oxide 두께를 줄인 결과, 좀 더 높은 민감도를 얻을 수 있었으며, 드레인 전압이 증가할 경우 민감도가 나빠진 다는 것을 확인하였다. 그리고 complete charge sheet model을 이용하여 underlap MOSFET의 모델링 결과도 얻을 수 있었다.
이러한 underlap을 이용한 바이오 센서의 경우 기존 FET 바이오 센서와 비교하여 높은 민감도, 간단한 공정과정, 기존 CMOS 공정과 주변회로와의 호환성이 좋다는 장점을 가진다.
