This research presents a MEMS based two-dimensional lens scanning module for forward endoscopic imaging. Two-dimensional forward scanning with high resolution is demonstrated implementing MEMS scannersof an x-scanner (in-plane comb drive) and y-scanner (vertical comb drive) together with a millimeter aspheric glass lens.
A light delivered from an optical fiber is coupled with a collimating lens and scanned with two scanning lenses, which has diameter of 1mm. Then it focuses to the image plan using an objective lens. To identify the feasibility of high resolution, optical simulations with a commercialized ray tracing program $ASAP^{TM}$ for beam divergence are performed. Simulation results present no significant change of the beam spot size until the light goes through inside of the ideal optical surface region of the objective lens. This represents about 3 degree of the scanning angle with an objective lens, which has 2mm diameter. The operation of each scanning lens is achieved by electrostatic MEMS actuators integrated on a chip with specially designed lens holders to grab lenses. Each scanner is designed to have maximum scan length of $\pm$40$\mum$ with considerations of simulation results.
Designed two-dimensional MEMS scanners are integrated to a micro optical bench with a special groove for optical fiber, considering both optical and mechanical requirements.
Micro optical bench is fabricated by standard SOI (Silicon on Insulator) wafer process, and then released with vapor phase HF gases so that the batch process at a wafer level is achieved.
Fabricated device is separated from wafer using specially designed fused tether technique due to the complex geometry of the micro optical bench. Then, the device eventually packaged on the Printed Circuit Board and glass lenses are mounted using UV curable epoxy resins.
Resonant frequency of the device is 2.1kHz before the lens mounting, however, decreases to 276.5Hz due to the heavy mass of the lens while, on the other hand, the quality-factor (Q-factor) is increased by a factor of 8.
Due to the high Q-factor gain by the lens mass, before the lens mounting, scanning angle of the device is 1.6 degree with 20V of dc and 10V of ac bias while scanning angle 3 degree is achieved under 5V of dc and 10V of ac bias after the lens mounting. The measurement result of beam divergence provides no reduction of optical resolution in scanning angle range of 3 degree. Finally, forward two-dimensional Lissajou`s pattern has been demonstrated with 3.6mm x 3.6mm scanning area.
In conclusion, two-dimensional MEMS lens scanning module is very functional for developing forward imaging system capable of fitting in an endoscopic capsule with optical biopsy schematics such as confocal and multi-photon imagings or optical coherence tomography.
광학적 생체조직검사(optical biopsy) 방법은 물리적인 절개 없이 질병을 진단 할 수 있어 기존의 조직검사를 대체할 수 있는 방법으로 각광받고 있다. 하지만 암과 같은 신체 내 질병을 진단하기 위해서는 내시경 모듈로의 제작이 필수적이나 소형화의 어려움으로 인하여 실제 임상에 적용되기에는 한계가 있는 실정이다. 특히, 이러한 생체 내 이미징을 위한 내시경 모듈을 제작함에 있어 조직 표면 위에서 빠른 속도로 광 주사를 구현하는 기술이 핵심이다. 이를 구현하기 위하여 MEMS 기술을 응용한 내시경 카테터의 연구가 최근 발표되고 있으나, 광 주사 방식을 2차원 거동이 가능한 미세거울에 의존하고 있어 측면 부 이미징만 가능하다. 뿐만 아니라 미세거울이 움직이면서 생기는 빛 번짐 (beam divergence) 현상으로 인하여 분해능 (resolution)의 한계가 있다. 이에 본 연구에서는 고분해능 전면부 이미징을 위해 MEMS 구동기 상에 미세렌즈를 집적하여 전면부 2차원 광 주사를 구현하였다.
본 연구를 통해 제작된 2차원 광 주사 모듈은 광섬유로부터 전달되는 광원을 직경 1mm 를 가지는 두 개의 미세렌즈를 통해 스캐닝 한 후 대물렌즈를 통해 샘플에 포커싱한다. 광학 수치해석 프로그램을 통해 분해능을 시뮬레이션 하였으며 주사되는 빛의 경로가 대물렌즈의 완전구면 영역을 지날 때 까지는 분해능의 저하가 없음을 확인하였다. 특히 직경 2mm 를 가지는 대물렌즈를 사용할 경우 스캐닝 각도 3도 이내에서는 분해능의 저하가 없었다. 이러한 광학 시뮬레이션 결과를 바탕으로 최대 $\pm$40$\mum$ 구동거리를 갖는 MEMS 구동기를 설계하였다. MEMS 구동기는 미세 콤 구조를 이용하여 정전기력을 이용하도록 설계하였으며, 미세렌즈를 집적하기 위한 보조 미세구조물들을 통해 미세렌즈를 스캐닝 할 수 있다.
본 연구를 통해 제작된 내시경 카테터는 2차원 광 주사를 위해 각 x 방향, y 방향으로 움직이는 2개의 MEMS 구동기 및 광 섬유를 집적하기 위한 구조물을 직경 3mm 이내의 미세광학벤치 상에 구현하였으며 광학적, 기계/기구적 및 전기적 요소를 동시에 만족할 수 있도록 설계하였다.
미세광학벤치는 SOI (Silicon on Insulator) 웨이퍼를 사용한 벌크 마이크로머시닝 (Bulk-micromachining) 과정을 통하여 제작되었으며 기체상태의 HF 가스를 이용하여 웨이퍼 수준에서 배치프로세스가 가능하게 하였다.
미세광학벤치의 복잡한 형상을 웨이퍼로부터 성공적으로 분리하기 위해 전기적으로 절단 가능한 실리콘 테더 (tether) 구조물을 사용하였으며, 제작된 미세광학벤치는 Printed Circuit Board (PCB) 기판 위에 패키징 한 후 자외선 경화수지 (UV curable epoxy resin) 를 이용하여 미세렌즈를 집적하였다.
제작된 소자의 공진 주파수는 미세렌즈의 집적 이전에 2.1kHz였으나 집적 이후에는 미세렌즈의 무게로 인하여 276.5Hz로 감소하였다. 반면 quality factor (Q-factor) 의 경우 공진주파수가 감소함에 따라 증가하여 약 8배 가량 증가함을 확인하였다. 이로 인해 미세렌즈의 집적 이전에는 직류전압 20V, 교류전압 10V 하에서 1.6도의 스캐닝 각도를 얻을 수 있었지만 미세렌즈의 집적 이후 직류전압 5V, 교류전압 10V 하에서 3도의 스캐닝 각도를 얻을 수 있었다. 빛 번짐 (beam divergence) 현상의 측정 결과 3도 이내의 스캐닝 각도에서는 분해능의 감소가 없음을 확인하였고, MEMS 구동기를 공진주파수에서 구동하여 3.6mm x 3.6mm 영역에서 리사쥬 패턴을 구현하였다.
본 연구를 통해 제작된 광 주사모듈을 사용하여 전면부 2차원 광 주사를 구현하였으며, 이는 공초점이미징법 (confocal imaging), 다중광자이미징법 (multiphoton imaging), 광결맞음단층이미징법 (Optical Coherence Tomograph: OCT) 등과 결합하여 광학적 생체조직검사를 위한 내시경 모듈에 적용될 수 있을 뿐 아니라 초소형 프로젝션 모듈 등 휴대용 소형광학시스템에도 적용 가능할 것으로 기대된다.
