Optical coherence tomography (OCT) was demonstrated as a promising imaging embodiment that enables high-resolution, cross-sectional noninvasive tomographic image. This technique is analogous to ultrasound imaging, except that it measures the reflected intensity of near-infrared (NIR) light, rather than sound waves. In optical coherence tomography based on the principle of the Michelson interferometer, the backscattered or back-reflected lights from different depths of the specimen are interfered with known delay light from the reference mirror. And functional imaging techniques in optical coherence tomography are based on the characteristics of the light such as polarization, Doppler Effect and absorption, which can give crucial information in some applications including the pathology and material analysis.
Spectral domain optical coherence tomography (SD-OCT) is usually composed of broadband light source, sample arm, reference arm and spectrometer for detection. It can achieve cross-sectional image of the internal micro-structures by Fourier-transforming the spectral interference pattern measured by the spectrometer. The polarization-sensitive spectral domain optical coherence tomography (PS SD-OCT) usually needs two detection channels in the spectrometer to detect two orthogonally polarized interference beams. This two-camera system has a high cost, a complex configuration, alignment errors and difficulties in synchronized triggering of two-camera spectrometer. Thus, the detection schemes based on single camera spectrometer have researched a lot.
In this thesis, polarization-sensitive spectral domain optical coherence tomography using multi-line single camera spectrometer is proposed. The novel spectrometer was based on the customized multiplexed grating and a three-line single camera. The incidence angles of two collimators are horizontally about an angle of $\plusmm 60$ degrees with regard to the multiplexed custom grating. After two polarized interference beams transmitted the multiplexed grating, they were focused on the different lines of the CCD camera by the camera lenses because the incident angles of two polarization channel beams have a small difference vertically. The center-to-center spacing between the two lines is $112 \mum$. Thus, without any time delay between them, two orthogonally polarized beams were simultaneously measured by a control signal of multi-line CCD camera. And the proposed system does not have any limitation of the camera full imaging speed and maximum depth range determined by the spectrometer resolution.
Spectral domain optical coherence tomography utilized a broad lighter (Broad lighter D830-HP1, Superlum Ireland) which is very low coherent light. Two SLD modules have different emission spectra each other and their total output power is around 7.82 mW. Thanks to the broadband light source with central wavelength of 826.4 nm and FWHM bandwidth of 84.7 nm, the axial resolution can be below $4 \mum$ theoretically. The laser beam was horizontally polarized by a linear polarizer. A quarter wave plate in reference arm is implemented to supply same reference power into both detection channels, with a linearly 45 degree polarization state with same amplitude and phase. Another quarter-wave plate is also implemented to illuminate the circular polarized probe beam into the sample. And the novel spectrometer of spectral domain optical coherence tomography was a homemade one which included a three-element air-spaced camera lens (f=120 mm, composed of a commercial NIR achromatic doublets and two designed singlet lenses) and a three-line CCD camera with 2098 elements ($14 \mum \times 14 \mum$) that can achieve 9.2k A-scan rate. In addition, the polarization state of the proposed system was analyzed by using the Jones matrix formalism and $Poincar\acute{e}$ sphere.
The Michelson interferometer and the novel spectrometer of polarization-sensitive SD-OCT were implemented on an anti-vibration table. The acquisition software was written by the custom-made Visual C++ program. Two dimensional raw data of two orthogonal interference spectra were saved onto a hard disk at a rate of about 8.98 frames per second ($2048 \times 1024$ pixels/frame) by the high-speed frame grabber. One axial scan time corresponding to the camera exposure period was $108.7\musec$. The post-processing was achieved by the Matlab and Labview program. To improve the accuracy of the polarization-sensitive image, zero padding technique and only data processing step above a certain threshold level were included. The fast real FFT to shorten the data processing time was applied to post processing part when the iterative dispersion compensation was needed.
The axial resolution was evaluated experimentally. The FWHM value of S-polarized beam is $3.8 \mum$, while its value of P-polarized beam is $4.5 \mum$. The depth profiles of two polarization detection channels, two axial peaks are almost matched. The objective lens with focal length of 19 mm gives the lateral resolution of about $10 \mum$ in air. The maximum imaging depth range is 1.9 mm in air due to the resolution of the spectrometer 0.09 nm. In this case, the maximal field of view (FOV) was around $2.5 mm (H) \times 1.9 mm (V)$. The SNR at optical depth $200 \mum$ was about 50 dB, but the SNR is decreased by 15 dB at 1mm distance due to finite pixel resolution in the spectrometer.
The reflectivity, single pass phase retardation and fast axis orientation images of the biological samples were obtained. The phase retardation and fast optic axis orientation images were encoded as color scale from 0 to 90 degree and from 0 to 180 degree, respectively. These were mouse tail tendon, human palm and mouse muscle including cancer, etc. The polarization-sensitive images of tendon mainly composed of collagen fiber demonstrate clearly the presence of the birefringence. Also, the phase retardation image of the human palm illustrates the presence of the collagen substances in dermis layers. The birefringence in normal mouse muscles of the right thoracic limb is convinced by the banded structures in phase retardation or fast axis orientation images. The heat treatment on the muscle deformed the banded structures in the polarization-sensitive images. And the normal chest muscle and LLC cancer of Ds red GFP mouse were discriminated by polarization-sensitive imaging. The difference between two samples was also confirmed in reflection and fluorescence images of confocal laser scanning microscopy.
광결맞음 단층촬영장치(OCT)는 고분해능의 비접촉 단면 영상을 획득 가능케 하는 유망한 이미징 수단으로 사용되어 오고 있다. 이 장치는 광원으로 음파가 아닌 근적외선을 사용하는 것이 다르지만, 초음파 영상 기술과 비슷한 점이 많다. 광결맞음 단층촬영장치는 보통 마이켈슨 간섭계 구조를 가지며, 기준 빔과 측정 대상물의 여러 깊이에서 되돌아오는 역산란 광이 간섭될 때, 깊이에 따른 지연시간을 측정하여 조직 내부의 영상을 획득하게 된다. 또한 광결맞음 단층촬영장치는 반사된 빛의 세기뿐만이 아니라 편광, 도플러 현상, 흡수 등을 이용한 기능적 이미징을 통해 병리학이나 물질 분석 등의 여러 분야에 중요한 정보를 줄 수 있다.
스펙트럼 영역의 광결맞음 단층촬영장치(SD-OCT)는 분광기에 의해 측정된 간섭 신호를 푸리에 변환을 통해 획득한 광축 방향 정보를 이용해 시편 내부의 절편 이미지를 획득하게 된다. 이 장치는 일반적으로 광대역의 스펙트럼을 갖는 레이저와 샘플 암, 기준 암, 간섭 신호 측정을 위한 분광기 등으로 이루어져 있다. 특히 편광에 민감한 스펙트럼 영역 광결맞음 단층촬영장치는 보통 서로 수직으로 편광 된 두 개의 간섭 빔을 이용하기 때문에 두 개의 측정 채널이 필요하다. 두 대의 카메라를 이용한 측정 방법의 경우 가격이 비싸지며, 그 구조가 복잡하며, 두 카메라 사이의 정렬이 어렵고 측정 시간 동기화가 쉽지 않아, 최근에는 한 개의 카메라로 구성된 분광기를 이용하는 연구가 많이 진행되어 오고 있다.
본 논문에서는 멀티라인 싱글 카메라 분광기를 이용한 편광에 민감한 스펙트럼 영역 광결맞음 단층촬영장치를 제안하였다. 이 분광기 시스템을 사용하면 카메라의 최대 측정 속도를 이용할 수 있으며, 분광기의 분해능에 의해 결정되는 최대 측정 깊이도 제약 없이 모두 이용할 수 있게 된다. 이 새로운 분광기는 특별히 주문 제작된 멀티플렉스 회절격자, 카메라 렌즈와 세 개의 측정 라인이 있는 카메라로 이루어져있다. 두 개의 시준기에 의해 서로 수직으로 편광 된 두 간섭 빔이 회절격자의 표면에 수평방향으로 각각 $\plusmm 60$도의 각도를 가지고 입사된다. 한편, 지면에 수직한 방향으로는 두 간섭 빔은 약간의 차이를 갖고 다른 각도로 입사된다. 그리하여 설계된 카메라 렌즈를 통과한 두 편광 간섭 빔은 $112 \mum$ 떨어진 두 픽셀 라인에서 각각 독립적으로 측정된다. 이 경우 두 편광 간섭 빔은 시간 지연 없이 하나의 카메라 구동 신호에 의해 동시에 측정된다.
본 연구에 사용된 광원은 두 개의 SLD 모듈이 결합된 저간섭성 광원으로써, 약 7.82 mW의 광 파워, 826.4 nm의 중심파장 그리고 84.7 nm의 밴드 폭(FWHM )을 갖는다. 따라서 이론적인 광축 방향 분해능은 공기 중에서 $4 \mum$ 이하 값을 갖는다. SLD 레이저에서 나온 빔은 선형 편광기에 의해 수평방향으로 선 편광 상태가 된다. 기준 거울에서 되돌아 오는 빛은 수평 방향에 22.5도의 각도로 설치된 quarter wave plate (QWP)에 의해 수평과 수직 방향의 편광 성분의 크기와 위상이 같은 45도의 선 편광 상태가 된다. 한편, 시편에 원평 편광 빔을 조사하기 위해 샘플 암에는 수평 방향에 대해 45도의 각도로 또 다른 QWP가 설치되었다. 본 연구에서 제작된 분광기는 주문 제작된 멀티플렉스 회절격자, 광학 설계 프로그램인 ZEMAX로 설계된 카메라 렌즈 그리고 Basler 사의 멀티라인 카메라로 구성되었다. 카메라 렌즈는 세 개의 렌즈로 구성되어 120 mm의 유효초점 거리를 가지고, Silicon 센서 기반 CCD 카메라는 최대 9.2 kHz로 작동되며 각각 2098개의 픽셀($14 \mum \times 14 \mum$)로 이루어진 세 개의 측정 라인을 갖고 있다. 또한 마이켈슨 간섭계에서의 빛의 편광 상태는 Jones matrix formalism과 $Poincar\acute{e}$ sphere를 이용하여 분석되었다.
제안된 측정 시스템은 제진 성능이 좋은 광 테이블 위에 설치되었다. 측정 소프트웨어는 크게 신호 획득 단계와 신호 처리 단계의 두 부분으로 나누어졌다. 신호 획득 프로그램은 Visual C++ 로 작성되었으며, 서로 수직으로 편광 된 두 간섭 빔에 대한 이차원 이미지 신호는 고속의 프레임 그래버에 의해 8.98 frames per second ($2048 \times 1024$ pixels/frame)의 속도로 하드디스크에 저장되었다. 하나의 측정 지점에 대한 광축 방향 주사 시간은 카메라 최소 노출 시간인 $108.7 \musec$ 이었다. 신호 처리 프로그램은 Matlab과 Labivew로 작성되었으며, 편광에 민감한 이미지인 phase retardation 이나 cumulative fast axis orientation의 정확한 측정을 위해 zero padding 기법이 포함되었고 특정 threshold 이상의 신호만이 처리되었다. 또한 반복적인 분상 보상이 필요한 경우 측정 시간의 절약을 위해 fast real FFT 알고리즘이 적용되었다.
광축 방향 분해능은 샘플 암의 편광 상태를 기준 암과 같게 한 후 실험적으로 평가되었는데, S 편광 간섭 빔과 P 편광 간섭 빔에 대한 분해능은 각각 $3.8 \mum$와 $4.5 \mum$이었다. 횡방향 분해능은 19 mm의 초점거리를 갖는 대물렌즈 사용시 약 $10 \mum$ 이었다. 최대 측정 가능 깊이는 분광기의 분해능 0.09 nm에 의해 공기 중에서 1.9 mm 로 결정되어, 측정 가능한 범위 (field of view)는 $2.5 mm (H) \times 1.9 mm (V)$이었다. 제안된 OCT 시스템의 신호 대 잡음 비 (SNR) 값은 광경로차 $200 \mum$에서 약 50 dB 이었는데, 분광기의 유한한 픽셀 분해능으로 인해 1 mm 지점에서 15 dB 정도 감소하였다.
다양한 복굴절 특성을 갖는 바이오 시편들의 reflectivity, single pass phase retardation 그리고 cumulative fast axis orientation 등이 제안된 시스템에 의해 측정되었다. Phase retardation 과 cumulative fast axis orientation 이미지는 각각 0에서 90도 그리고 0에서 180도 사이의 값을 갖는데, 이는 색깔로써 표시되었다. 주요 측정 시편은 쥐꼬리의 힘줄 (tendon), 사람 손바닥 피부, 쥐의 다리 및 가슴 근육 및 암세포 (LLC cancer) 등이다. Phase retardation image 를 통해 쥐 꼬리 힘줄의 복굴절 (birefringence) 특성을 확인하였으며, 사람 손바닥의 진피 층에서부터 복굴절 특성이 나타났는데 이는 collagen 성분으로 인한 것이다. 또한 쥐 다리 근육에서 복굴절 특성으로 인한 줄무늬 구조가 phase retardation 과 fast axis orientation 이미지에 나타났으며, 열로 인한 변형도 제안된 시스템으로 확인할 수 있었다. 또한 쥐의 가슴 부위에 있는 정상 근육과 이 곳에 생긴 암세포의 차이는 편광에 민감한 OCT 이미징을 통해 분별되었다. 암세포와 정상 가슴 근육의 이미지를 자체 제작된 반사 형광 복합 공초점 현미경로 획득 했을 때도 유사한 측정 결과를 보였다.
