Surface modification of substrates has been widely implemented in various areas including materials sciences, nanotechnology, and biotechnology. In particular, encapsulation of individual nanoparticles, surface modification for nanoparticles, has gained great attention as a method for both stabilizing nanoparticles and tailoring surface properties of nanoparticles. Especially, the encapsulation of nanoparticles with silica shells is advantageous for bioconjugation and applications to (nano)biotechnology. In chapter 1 and 2, we reported a method for constructing gold nanoparticle (AuNP)/silica core/shell hybrid structures by biomimetic silicification of silicic acids. The procedure in chapter 1 consisted of surface-initiated, atom transfer radical polymerization of 2-(dimethylamino)ethyl methacrylate (DMAEMA) from AuNPs and biomimetic polycondensation of silicic acids by using poly(DMAEMA) as a synthetic counterpart of silaffins that were found in diatoms. The resulting AuNP/silica hybrids were characterized by Fourier-transform infrared spectroscopy, energy dispersive X-ray spectroscopy, UV-vis spectroscopy and transmission electron microscopy. In addition, the immobilization of biological ligands onto the hybrids was investigated for potential applications to biotechnology. As a model ligand, biotin was attached onto the AuNP/silica hybrids through substitution reaction and Michael addition reaction, and the attachment was confirmed by fluorescence microscopy after complexation with fluorescein-conjugated streptavidin. In chapter 2, we formed ultrathin (~1.5 nm) silica shell-coated nanomaterials by using self-assembled monolayers as organic templates for silicification. The procedure consisted of simple two steps: immobilization of a thiol-terminated imidazolium ion onto AuNPs and biomimetic polycondensation of silicic acids. The resulting AuNP/silica hybrids were characterized by Fourier-transform infrared spectroscopy, transmission electron microscopy, and UV-vis spectroscopy.
Chapter 3 describes a method for silica encapsulation of magnetic nanoparticle. We extended our strategy to magnetic nanoparticles for silica coating through biomimetic approach. DMAEMA was grafted onto the surface of IONPs via surface-initiated, atom transfer radical polymerization, and then silica shell was formed by biomimetic polycondensation of silicic acids with poly(DMAEMA) as an organic template for forming silica structures.
In addition to silica encapsulation of nanoparticle, substrate-independent silica coating method was also investigated. In chapter 4, we reported a biomimetic method inspired by mussel and diatom to form silica thin films onto various types of materials, including metal oxide and polymers. First, we formed functional surfaces on the substrates using self-polymerization of 2-(3,4-dihydroxyphenyl)ethylamine (dopamine) and subsequent modification with amine compound. After that, silica thin films were fabricated along the amine-functionalized polydopamine surfaces.
Chemical modification of $Ti/TiO_2$ substrates has recently gained a great deal of attention because of the applications of $Ti/TiO_2$-based materials to biomedical areas. The reported modification methods generally involve passive coating of $Ti/TiO_2$ substrates with protein-resistant materials, and poly(ethylene glycol) (PEG) has proven advantageous for bestowing non-biofouling property on the surface of $Ti/TiO_2$. However, it is required to introduce biologically active moieties onto $Ti/TiO_2$, in addition to non-biofouling property, for the wider applications of $Ti/TiO_2$-based materials to biomedical areas. In chapter 5 and 6, we utilized surface-initiated polymerization to coat $Ti/TiO_2$ substrates with polymers presenting the non-biofouling PEG moiety and subsequently conjugated biologically active compounds to the PEG-presenting, polymeric films. Specifically, a $Ti/TiO_2$ surface was chemically modified to present an initiator for atom transfer radical polymerization, and poly(ethylene glycol) methacrylate (PEGMA) was polymerized from the surface. After activation of hydroxyl groups of poly(PEGMA) (pPEGMA) with N,N’-disuccinimidyl carbonate, biotin and BMP-2, model compounds, were conjugated to the pPEGMA films, respectively. The reactions were confirmed by infrared spectroscopy, contact angle goniometry, and ellipsometry. The biospecific binding of streptavidin was also utilized to generate micropatterns of proteins on the $Ti/TiO_2$ surface, and the increased bioactivity, which was induced by BMP-2, of $Ti/TiO_2$ was proved by checking the cellular response of mesenchymal stem cells on the $Ti/TiO_2$ substrates.
Chapter 7 describes a method for chemical modification of magnetic nanoparticles (MNPs) with functional polymers. Chemical modification of MNPs with functional polymers has recently gained a great deal of attention because of applications of MNPs to in vivo and in vitro biotechnology. Especially, the potential use of MNPs as capturing agents and sensitive biosensors has intensively been investigated, because MNPs would show good separation-capability and binding-specificity for biomolecules after suitable surface functionalizations. We demonstrated an efficient method for surface modification of MNPs, which combined surface-initiated polymerization and subsequent conjugation of biologically active molecules. The polymeric shells of non-biofouling pPEGMA were introduced onto the surface of MNPs by surface-initiated, atom transfer radical polymerization. With biotin as a model of biologically active compounds, post-functionalizations of the polymeric shells-activation of the polymeric shells and bioconjugation of biotin-were successfully performed. The resulting MNP hybrids showed the biospecific binding property for streptavidin and could be separated by magnet capture.
최근 계면에서 일어나는 현상에 대한 연구가 활발히 이루어지고 있는데, 이는 몸 속에서 일어나는 반응들이 대부분 용액 중에서 일어나는 것이 아닌 계면에서 일어나기 때문이다. 따라서 기초과학 분야뿐만 아니라 의학분야에 이르기까지 계면 현상에 대한 연구가 활발히 진행되고 있다. 특별히 계면현상에 관한 연구를 진행할 때에는 물리적 차원에 따라 2가지 접근이 가능한데, 2차원 기판과 3차원 기판에 대한 연구가 바로 그 것이다. 여기서 말하는 2차원 기판은 흔히 plate 형상의 고체기제를 말하는 것이고, 3차원 기판은 입자 형태의 고체기제를 말한다. 본 연구에서는 먼저 3차원 기판의 한 형태인 나노입자의 표면 성질을 제어하기 위한 연구를 수행하였다. 나노입자의 표면 개질에 있어서는 최근 실리카를 이용한 껍질형성 연구가 활발히 이루어지고 있다. 즉, 우리가 원하는 나노입자를 중심에 두고 실리카를 이용하여 껍질을 형성시키는 것인데, 응용 연구에 있어서 안정도 증대가 필요시 되는 나노입자를 실리카 껍질로 코팅시켜줌으로써 안정도를 높여줄 수 있다. 실리카 코팅은 비단 안정도를 높여줄 뿐만 아니라 현재까지 잘 정립되어온 실리카 화학을 이용하여 원하는 물질을 나노입자 표면에 고정시킬 수 있다는 장점도 지니고 있다. 본 연구에서는 나노입자의 표면을 실리카 껍질로 코팅하기 위한 방법으로 생체모사 규화반응을 이용하였다. 생체모사 규화반응은 현재까지 알려진 화학적 규화반응에 비해 온화한 조건에서 짧은 시간안에 실리카 코팅을 할 수 있는 방법으로써 최근 연구되고 있는 반응이다. 이러한 생체모사 규화반응을 금 나노입자 및 산화철 나노입자에 적용하였으며 그 결과, 실리카로 균일하게 코팅된 나노입자/실리카 복합체를 형성시킬 수 있었다. 또한 이렇게 형성된 나노입자/실리카 복합체의 표면을 실란 화합물을 이용하여 한번 더 개질함으로써 복합체의 응용가능성을 보였다. 생체모사 규화반응에 있어서 2차원 기판에서도 연구를 진행하였는데, 현재까지 보고된 표면 위에서의 생체모사 규화반응의 표면 제한성을 보완하고자 하였다. 지금까지는 생체모사 규화반응을 이용하기 위해 표면에 따른 적당한 화합물질을 선정하여 먼저 코팅하는 작업을 거쳤으나, 최근 보고된 표면-무관성 코팅방법을 이용하여 어떠한 표면이든 쉽게 생체모사 규화반응을 적용시킬 수 있도록 하였다.
생체모사 규화반응과 더불어 생체물질의 비특이적 흡착 방지 표면에서의 선택적 생체물질 고정화에 대한 연구도 진행하였다. 이 연구는 특별히 최근 의학용으로 많은 관심을 받고 있는 재료인 티타늄과 자성 나노입자에 대해서 진행하였다. 티타늄의 경우 그것이 갖는 생체적합성 및 높은 강도로 인해 임플란트의 재료로서 각광을 받고 있다. 하지만 티타늄을 고효율의 임플란트 재료로 사용하기 위해서는 해결해야 할 점이 있다. 티타늄이 높은 생체적합성을 갖고는 있지만 생체 활성도가 낮기 때문에 티타늄 주변으로의 골 형성율이 좋지 않은 점이 바로 그 것이다. 이러한 문제를 해결하기 위해 티타늄의 표면에 생체활성물질을 선택적으로 고정하고자 하였다. 생체활성물질을 고정할 때에는, 임플란트가 생체 내부에서 쓰일 때 야기될 수 있는 문제점인 비특이적 흡착을 배제하기 위해 비특이적 흡착 방지 표면처리를 한 뒤 수행하였는데 표면 개시 고분자 중합법을 이용하여 표면 처리하였다. 생체활성물질로는 streptavidin과 선택적 결합을 하는 것으로 알려진 biotin과 골 형성 단백질(BMP-2)를 사용하였다. 실험 결과 biotin이 고정된 티타늄 기판에는 streptavidin만이 흡착을 일으켰으며, 원하지 않는 단백질인 fibrinogen, lysozyme, bovine serum albumin 등은 전혀 흡착을 일으키지 않음을 확인할 수 있었다. 또한 BMP-2가 고정된 티타늄 기판의 생체활성을 확인하기 위해 줄기세포 분화실험을 진행하였다. 두 번째로 사용한 생체활성물질인BMP-2는 줄기세포가 표면에 잘 접합하도록 돕는 역할을 할 뿐만 아니라 줄기세포가 골아세포로 분화되도록 돕는 역할을 한다. 줄기세포 실험 결과, BMP-2가 고정되지 않은 비특이적 흡착방지 처리된 티타늄 표면에서는 세포가 전혀 접합되지 않는 반면, BMP-2가 고정된 기판에서는 줄기세포가 잘 접합될 뿐만 아니라 골아세포로 분화되는 것을 확인할 수 있었다. 이를 통해 생체활성물질을 티타늄 기판에 효과적으로 고정화시킬 수 있었다.
마지막으로 자성 나노입자의 표면에 선택적으로 생체물질을 고정화하는 연구도 함께 수행하였다. 앞에서 언급한 티타늄 연구와 마찬가지로 표면 개시 고분자 중합법을 사용하여 자성 나노입자의 표면개질을 시도하였으며, 이를 통해 먼저 생체물질의 비특이적 흡착 방지 표면을 형성하였다. 이러한 표면 위에 biotin을 고정한 뒤 streptavidin 용액과 함께 섞어주면 biotin이 고정되어 있는 자성 나노입자와 streptavidin이 복합체를 이루게 되고, 외부에 자석을 가까이 접근시킴으로써 streptavidin을 선택적으로 분리할 수 있다. 이러한 연구는 자성 나노입자의 표면에 고정시키는 생체물질을 달리함으로써 특정물질의 분리를 위한 목적으로 사용할 수 있다.
