RhoB plays a role as a tumor suppressor, causing growth-inhibitory and apoptotic effects on cancer cells and also known to be a critical mediator for the apoptosis by many anti-cancer agents.
Here, we report a novel piperazine alkyl derivative, KHG2 for an anti-cancer drug, which upregulate RhoB expression, leading to apoptosis in human cancer cells. KHG2 showed a good cytotoxicity toward human cancer cells in-vitro and mouse xenograft assay using NUGC-3, a human stomach cancer cell line. KHG2 induced a G2-M arrest and apoptosis in NUGC-3. KHG2 activated JNK pathway, thereby increasing phospho-c-jun in NUGC-3. And, prior treatment of a JNK inhibitor, SP600125, inhibited most of the apoptosis, suggesting that the activation of JNK might play a crucial role for the apoptosis by KHG2. Furthermore, whereas KHG2 decreased phospho-forms of both PTEN (Ser380) and Akt (Thr308) as well as total amounts of FTase-1α and K-ras, it dramatically increased p-Bcl2 (Ser70) and RhoB. The functional role of RhoB was proved by ectopic expression of the gene in NUGC-3 that induced the eminant apoptosis. These results clearly show that a novel compound, KHG2, induces the anti-cancer activity via dysregulation of the multiple pathways including activation of JNK and RhoB, thereby inducing apoptosis in NUGC-3.
In spite of the many previous reports of anti-cancer activities of RhoB, the exact mechanisms for the apoptosis by RhoB are not proved well. Here, we report that TNFAIP1 induces an apoptosis through direct interaction with RhoB in Hela, a human cervical cancer cell line. Through the yeast-two hybrid screening using Hela cDNA library as a prey, we found many interesting proteins including TNFAIP1. RhoB also interacted with TNFAIP1 in-vitro and In-vivo, Furthermore, co-transfection of both RhoB and TNFAIP1 induced eminent apoptosis compared to single transfection. And RhoB knock down using RhoB-specific si-RNAs inhibited the apoptosis by TNFAIP1 in Hela cells, suggesting that direct interaction with RhoB is critical for the apoptosis by TNFAIP1 in Hela cells.
Ras-homolog member B (RhoB) 는 a tumor suppressor로 작용하여 암세포의 성장 억제와 apoptosis를 유도한다. 또한, 여러 가지의 항암 치료제의 apoptotic mediator로서 알려져 있으며, 새로운 cancer drug target으로서 떠오르고 있다. 이러한 관점에서 RhoB를 타겟으로 하는 새로운 약물의 발견은 새로운 항암제 개발 외 에도, RhoB의 작용 기전을 규명하는데 있어 중요하다고 할 수 있다. 따라서, 본 연구에서는 RhoB를 증가시키는 새로운 항암 약물로서의 piperazine alkyl 유도체인 KHG2를 발굴하였고, 이의 작용 기전과 RhoB의 역할을 규명하였다. KHG2는 in vitro, in vivo 상에서 좋은 항암 효과를 보여주었다. KHG2 는 위암 세포 주인 NUGC-3에서 G2-M arrest와 apoptosis를 유도하였고, 이 과정에서 JNK와 RhoB 활성화를 유도하였다. 이 외에도, p-PTEN (Ser380)과 p-Akt (Thr308) 및 FTase-1, K-ras 감소 및 p-Bcl2 (Ser70)의 증가를 통해 세포의 성장과 사멸에 중요한 pathway를 조절함을 확인 할 수 있었다. 특히, RhoB를 현격하게 증가시켰고, ectopic expression을 통해 RhoB가 NUGC-3 에서 현저하게 apoptosis를 유도함을 확인하였다. 이러한 결과들을 종합해보면, KHG2는 JNK 와 RhoB 활성화를 포함하여 여러 가지의 multiple pathways를 deregulation함으로써 항암 작용을 수행하리라 여겨진다. 특히, JNK와 RhoB의 활성화는 KHG2에 의한 apoptosis를 inducing하는데 중요한 매개체 역할을 하리라 여겨진다. RhoB는 많은 항암 작용에 대한 보고에도 불구하고 세포 사멸 유도 기작에 대한 이해는 아직도 미흡한 상태이다. 따라서, RhoB의 항암 기전을 규명코자 Hela cDNA library를 이용하여 yeast-two hybrid screening을 수행하였고 이를 동해 RhoB와 direct interaction하는 새로운 단백질로서 TNFAIP1을 identify하였고, in vitro binding assay 및 in vivo FLAG- immunoprecipitation assay를 통해 RhoB와 TNFAIP1의 interaction을 확인하였다. 또한, GFP-RhoB 혹은 GFP-mDia1과 RFP-TNFAIP1을 co-transfection시킨 후, 형광 현미경을 이용한 co-localization study를 통해, 이들 단백질들이 세포질 내의 endosome에서 interaction하는 것으로 여겨진다. TNFAIP1은 TNF에 의해 induced 되어지는 것으로 보고되었고, apoptosis를 야기하는 것으로 여겨지나, 자세한 작용 기작에 대한 보고는 전무한 상태이다. 본 연구에서는 transfection study를 통해 TNFAIP1이 자궁 경부암 세포 주인 Hela cell에서 apoptosis를 유도함을 확인하였고, RhoB와 co-transfection시켰을 때 현격하게 apoptosis를 증진시킴을 확인하였다. 이 과정에서 JNK활성화가 중요함을 확인하였다. 특히, si-RhoB RNAs를 통해 RhoB를 knock-down 시켰을 때 TNFAIP1에 의한 세포 사멸이 크게 줄었다. 이는 RhoB의 존재가 TNFAIP1에 의한 apoptosis에 중요함을 의미한다. 결론적으로, 우리는 본 실험 결과를 토대로 RhoB가 신규 항암 약물인 KHG2와 TNFAIP1에 의한 암 세포의 사멸에 중요한 매개체임을 제안했다.
