The role and molecular mechanisms of a new Hippo signaling pathway are not fully understood in mammals. Here, we generated mice lacking WW45 and reveal a crucial role of WW45 in cell-cycle exit for epithelial terminal differentiation. Many organs in mutant embryos displayed hyperplasia accompanied by defects of terminal differentiation of epithelial cells owing to impairment of proliferation stop of epithelial cells rather than intrinsic acceleration of proliferation during differentiation. Importantly, MST1 signaling pathway is specifically activated in only differentiating epithelial cells. We also found that WW45 is required for MST1 activation and translocation to the nucleus for subsequent LATS1/2 activation, which then phosphorylate serine 127 of YAP upon differentiation signal. This phosphorylated YAP translocates from the nucleus into the cytoplasm, thereby resulting in cell-cycle exit and terminal differentiation. Collectively, these data provide compelling evidence that WW45 is a key mediator of MST1 signaling in the coordinate coupling of proliferation stop with terminal differentiation for proper epithelial tissue development in mammals.
조상세포 (progenitor cell)는 정상적인 상피 발생에서 일정 횟수의 세포 분열을 마친 뒤 최종 분화하게 된다. 이러한 조상세포의 세포 분열과 분화의 균형이 망가질 경우 암이 발생하기도 한다. 최근 초파리에서 기관 발생시 세포 분열과 사멸을 동시에 조절하는 새로운 신호전달 경로인 ‘Hippo pathway’ 가 알려졌다. ‘Hippo’, ‘Salvador’ and ‘Warts’ 라는 단백질 복합체를 주축으로 한 kinase cascade로 하위 전사 조절 단백질인 ‘Yorkie’를 억제함으로써 세포 주기 조절 단백질인 ‘Cyclin E’와 세포 사멸 조절 단백질인 ‘DIAP1’ 등의 단백질 발현을 조절한다. 포유 동물에서도 그 orthologue들이 존재하는 것으로 밝혀져 그들의 결합여부가 보고되고 있으나 아직까지 정확한 기능과 신호전달 경로의 분자적 기작이 연구되어 있지 않다. 따라서 본인은 포유 동물에서 초파리 ‘Salvador’의 유일한 homologue인 ‘WW45’의 유전자 적중 마우스를 제작하여 포유동물의 상피 발생에서의 이들의 기능을 연구하였다.
WW45 유전자 적중 마우스는 대부분의 상피 조직에서 과증식과 함께 미분화를 보이며 부분적 극성의 소실과 같은 전암 증상을 나타내었다. 더욱 흥미로운 것은 이러한 분열하는 세포들의 비대 증식이 최종 분화를 위한 세포 주기 조절의 실패로 기인한다는 것이다. 상피 세포들이 분화할 때 특이적으로 ‘Hippo’의 포유동물 orthologue인 ‘MST1/2’가 활성화 되는 것을 확인 하였고, WW45가 이렇게 활성화된 MST1/2를 세포질에서 핵으로 이동시켜 ‘Warts’의 포유동물 orthologue인 ‘LATS1/2’를 인산화 하여 활성화 시키고, 이것이 그 하위 단백질인 ‘YAP’, 즉 ‘Yorkie’의 포유동물 ortholoue의 serine 127 residue를 인산화 하여 핵 안에서 세포질로 이동시킴으로써 전사조절 기능을 억제 시킨다. 즉, 세포 분열이 멈추고 최종분화가 일어나 정상적인 상피 발생이 일어나게 된다. 이러한 연구 결과는 포유 동물의 상피 조직 발생에 있어서 ‘MST pathway’의 새로운 신호 전달 기작을 밝힌 중요한 증거이다. 또한, ‘MST signaling’이 세포 분열과 분화의 균형을 조절하는데 ‘WW45’가 중요 매개체 역할을 한다는 것을 생체 내에서 밝힌 최초의 연구 결과이다.
