The patterning of biomolecules at designated locations is essential for the fabrication of biologically multifunctional devices such as array-based biochips for diagnosis. It has been explored that biomolecules are spatially immobilized on electrochemically active layers. We prepared and characterized electrochemically active polymeric layers on ITO electrodes and found their applications to biomolecule patterning. Self-assembled polymeric layers on ITO coated glass were prepared in water from a series of random copolymers of 3-(trimethoxysilyl)propyl methacrylate (TMSMA), poly(ethylene glycol) methyl ether methacrylate (PEGMA) and p-aminophenol (p-Ap), denoted as poly(TMSMA-r-PEGMA-r-p-Ap). An electrochemical oxidation of the resulting polymeric layers enables thiol terminated molecules to be immobilized on the surface via 1,4-addition of thiols to p-Ap moieties. Analyses by cyclic voltammetry, x-ray photoelectron spectroscopy, silver enhancement, and 4-chloro-1-naphthol precipitation show that the resulting polymer modified ITO provide effective and selective surfaces for immobilizing alkanethiols, biotins and aptamers.
우리는 이번 연구에서 공중합체인 Poly(TMSMA-r-PEGMA-r-p-Ap)를 이용하여 산화인듐주석 전극 위에 자기 조립 단분자층을 형성시켰다. 이 공중합체의 trimethoxysilane 부분이 산화인듐주석과 결합한다. 공중합체의 p-Ap(p-aminophenol) 반응기를 전기화학적 전위를 줌으로써 산화시키고, cysteine, 1,6-hexanedithiol, biotin-HPDP와 같이 -SH 작용기를 가지고 있는 물질(thiol)이 친핵체로 작용하여 산화가 일어난 표면의 p-quinoneimine(p-Ap가 산화된 형태)과 결합하도록 하였다. 순환 전압전류법 (CV)을 사용하여 thiol을 표면에 고정화시킴과 동시에 상태 변화를 관찰할 수 있었다. 전기화학적인 방법 외에 엑스레이 광전자 스펙트로스코피 (XPS), silver enhancement 방법을 통해서 표면 개질을 관찰하였으며, 산화된 공중합체층과 산화되지 않은 공중합체층의 차이를 구별할 수 있었다.
공중합체 Poly(TMSMA-r-PEGMA-r-p-Ap)는 PEG(polyethylene glycol)를 많이 포함하고 있으므로, 단백질이 산화인듐주석 표면에 비특이적으로 달라붙는 것을 막아준다. 선택적으로 산화된 공중합체층에 biotin-HPDP와 -SH 말단을 가진 TBA(트롬빈 단백질에 선택적으로 결합할 수 있는 압타머)를 반응시켰다. 4-chloro-1-naphthol 침전 반응을 통하여 바이오틴과 압타머가 산화된 표면에 선택적으로 결합했음을 알 수 있었다.
