Ovarian cancer is one of the most lethal gynecologic malignancies. Since most ovarian cancer cases were diagnosed in the late stages, drug resistance and metastasis are the major impediment in successful treatment of ovarian cancer. It is now commonly accepted that aberrations in DNA methylation may play an important role in tumorigenesis and drug resistance. It was also suggested that DNA demethylating agents would be promising chemotherapies to reverse drug-resistant phenotypes in previous studies. To investigate the effect of DNA demethylating agents in resensitization, cytotoxicity assays and several apoptosis assays were performed using R5, the cisplatin-resistant ovarian cancer cell line which had been established in vitro. Pretreatment of 5-aza-dC or zebularine was shown to cause decrease of 50% effective concentration of cisplatin in dose-dependent manner. Although there were no clear evidences of cisplatin-induced apoptosis in both R0 and R5 cells, it can be suggested that combined treatment of 5-aza-dC or zebularine with conventional chemotherapies is worth to develop as a promising strategy for overcoming drug-resistant tumors.
난소암은 여성암 중에서 가장 사망률이 높은 질병으로, 초기 진단이 어려워 대부분 말기에 많이 발견이 된다. 이 경우 약제저항성이나 암전이 등으로 발전되는 경우가 많아 난소암 치료의 가장 큰 걸림돌이 되고 있다. 과거 많은 연구들을 통해 암의 진행과정에서는 물론 약제저항성을 획득하는데에 DNA 메틸화의 증가가 중요한 역할을 하고 있다는 것이 밝혀졌다. 따라서 DNA 메틸화를 억제하는 DNA 탈메틸화제가 약제저항성을 극복하는 효과적인 치료법으로서 새로이 대두되고 있다. 이에, 두 가지 DNA 탈메틸화제인 5-aza-dC와 zebularine이 난소암 치료에 쓰이는 항암제의 하나인 cisplatin에 저항성을 가진 난소암 세포주에서 약제감수성을 회복하는데 효과가 있는지를 조사해보았다. 비록 apoptosis assays를 통해 cisplatin에 의해 유도되는 세포사멸을 확인하지는 못하였으나, cytotoxicity assay는 5-aza-dC와 zebularine이 난소암세포의 약제감수성 회복에 어느 정도 효과가 있음을 보여주고 있다.
