The receptor tyrosine kinases, tie1 and tie2, which is largely endothelial cell specific, and angiopoietin-1, 2, secreted ligand for tie2, are recently discovered to be critically involved in the formation of vasculature. Tie receptor family consist of tie1 and tie2 that affect differently early development and tumor - induced angiogenesis. And ligands for tie2, termed angiopoietin-1, angiopoietin-2, also play crucial role in the maturation and maintenance of the mammalian vascular and lymphatic system. Angiopoietin ligands can function as agonist or antagonist in signal pathway of tie although the share significant amino acid sequence identity. A variety of disease is associated with abnormal expression of tie2, angiopoietin. For example, the insufficient formation of blood vessel causes heart disease, or stroke. On the other hands, deregulated blood vessel formation brings about pathogenesis of diabetic proliferative retinopathy, or solid tumor growth.
The ectodomain of tie2 is organized by three Ig domains, three epidermal growth factor, and three fibronectine type Ⅲ repeats. And the Ig- EGF region mediates angiopoietin recognition and binding.
Angiopoietins have an N terminus which is involved in superclustering, coiled- coil domain and a fibrinogen - like region at the C terminus. It is reported that N terminus and coiled- coil domain are essential for ligand oligomerization, while fibrinogen - like C terminus is concerned with the interaction of tie2 receptor. To determine the structure of tie2 angiopoietin complex, we have obtained the crystal of Tie2/Ang-1, Tie2/Ang-2 complex and collected x-ray diffraction data of Tie2/Ang-2 complex at a resolution of 2.9Å. However, we failed in improving crystal of Tie2/-1 complex in shapes and size.
We expect that our study can contribute to development of novel drug for disease related to blood vessel.
내피 특이성을 가지는 Tie2 수용체 tyrosine kinases 와 그에 대한 리간드로 알려진 angiopoietin (Ang)는 혈관생성에 중요한 영향을 끼치는 것으로 알려져 있다. 특히 발생학적 혈관 생성뿐 만 아니라 종양에 의해 유도되는 혈관생성이나 당뇨합병증, 심혈관계 질환에 중요한 역할을 하는 것으로 알려져 그 연구가 굉장히 중요하다고 할 수 있다. 이 외에도 혈액생성과 림프관 생성과 신경계와 안구 등 다양한 조직에서의 혈관생성에 깊이 관여하고 있는 것을 여러 연구 결과를 통해 알 수 있다. 그 중 Tie2 수용체에 대한 작용제로 알려진 Ang-1와 길항제로 알려진 Ang-2는 다양한 조직과 세포 내에서 적당한 조절을 통해 혈관의 정상적인 유지에 중요한 역할을 하고 있다. 특히 Ang-2는 VEGF의 농도나 Ang-1의 농도 등 다양한 상황에 따라서 작용제 혹은 길항제로 작용하여 혈관 생성에 관여하고 있다.
이러한 Ang-1와 Ang-2의 정확한 역할과 기능 연구를 위해서는 Tie2 수용체와의 복합체의 구조적인 연구가 필요하다. 이 논문에서는 Tie2 수용체의 extracellular domain과 Ang-1, Ang-2의 복합체의 구조를 crystallography를 통하여 규명하기 위해서 단백질 결정을 얻었다. 결정을 얻기 위하여 vaculovirus 체제를 이용, insect cell을 통해 단백질을 얻었으며, 다양한 정제 방법을 통해 깨끗한 상태의 단백질 복합체를 얻을 수 있었다. 단백질 복합체를 다양한 조건에서 결정화를 시도하여 방사형의 Tie2/Ang-1 복합체의 결정을, Tie2/Ang-2는 판형의 결정을 얻을 수 있었다. 다양한 조건에서의 결정화를 통해 결정의 모양이나 크기에의 개선을 시도하여, Tie2/Ang-2 복합체의 경우, 좀 더 단단한 형태의 판형의 결정을 얻을 수 있었다. 그러나 Tie2/Ang-1의 경우, X선 상에 회절 데이터를 얻을 수 있을 만큼의 좋은 결정으로의 개선을 이루지 못하였다. Tie2/Ang-1의 경우, 2.9Å의 X선 회절 데이터를 얻을 수 있었다. 이러한 회절 데이터를 이용한 단백질의 구조 분석을 통해 비정상적인 혈관에 의해 유도되는 암이나 당뇨합병증 등의 질환의 신약 개발에 유용하게 이용될 수 있다고 생각한다.
