Sodium/Hydrogen $(Na^+/H^+)$ antiporters, briefly SHA's, are ubiquitous membrane proteins that are involved in homeostasis of H+ and Na+ throughout the biological kingdom. Corroborating their roles in pH homeostasis, many of the SHA proteins are regulated directly by pH. The major objective of this study was to understand the roles of SHA's in the production and/or secretion of secondary metabolites by S. coelicolor A3(2) M145. For this purpose, sha 7 gene (SCO7832) was deleted from S. coelicolor A3(2) chromosome since it was one of the two sha genes, together with sha 4, that showed the strongest gene dosage effect on actinorhodin and undecylprodigiosin production. The transformant of Δsha 7 was constructed by double-crossover between recombinant plasmid pKC1139-LRNeo and the S. coelicolor A3(2) chromosome. When the initial pH and NaCl concentration were changed in the ranges of 4 ~ 7 and 0 ~ 1 M, Δsha 7 showed lower production of actinorhodin and undecylprodigiosin than the wild strain in overall, as expected. No notable pH effects were observed, while a high NaCl concentration reduced their production in Δsha 7 more severely than in the wild strain. When an acidic pH shock was applied to Δsha 7, it was observed that the secondary metabolites production was significantly suppressed in overall.
$Na^+/H^+$ antiporters 는 모든 생물계가 가지고 있는 막단백질로 $Na^+$와 $H^+$ 의 항상성을 조절한다. pH 조절하는 역할과 함께, 많은 $Na^+/H^+$ antiporters 은 pH 에 의해 조절을 받는다.
이 연구의 주된 목적은 S. coelicolor A3(2) M145 에 의해 생산되는 이차대사산물의 생산 혹은 분비와 관계 있는 Sodium Hydrogen Antiporter (SHA) 의 역할에 대해 이해하는 것이다. 이 목적을 위해, actinorhodin 과 undecylprodigiosin 생산에 있어 gene dosage effect가 가장 큰 유전자 중 하나인 (sha 4 와 함께) sha 7 유전자를 (SCO7832) S. coelicolor A3(2) 염색체로부터 제거하였다. 그리고 이렇게 만들어진 sha 7 돌연변이체와 야생형의 특성을 비교하고자 초기 pH 와 NaCl 농도에 따른 영향을 알아보았고, 돌연변이체는 산충격에 의해 어떻게 반응하는지도 알아보았다.
sha 7 돌연변이체는 S. coelicolor A3(2) 염색체와 pKC1139-LRNeo 재조합 플라스미드 사이에서 유전자가 같은 부분에서 double-crossover에 의해 얻어졌고, 이렇게 sha 7 이 제거된 돌연변이체의 확인은 PCR과 Southern hybridization 에 의해 최종 확인이 되었다.
TES 가 없는 SMMS 배지의 초기 pH를 4~7 로, NaCl 농도를 0~1M 로 만든 후, 6일간 배양하여 세포의 건조중량과 세포 내, 세포 밖의 actinorhodin 과 undecylprodigiosin 의 양을 측정한 실험에서, 예상했던 바대로 sha 7 돌연변이체는 야생형에 비해 전체적으로 두 항생제의 생산이 낮았다. 특히, 높은 NaCl 농도에서 sha 7 돌연변이체는 두 항생제의 생산에 있어 큰 저해를 받고 있었지만, 초기 pH 에 따른 어떤 경향성은 관찰할 수가 없었다. 또한, sha 7 돌연변이체에 산 충격을 가한 실험에서도, 두 항생제의 생산이 전체적으로 산충격을 가하지 않았을 때보다도 낮았다. 이것은 sha 7 이 제거되어 산충격을 가했을 때, 야생형에 비해서 pH가 정상적으로 회복이 되지 못했기 때문이라 추측된다. 이런 모든 결과들을 고려해 볼 때, sha 7 유전자는 pH 항상성과 두 색깔있는 항생제의 생산 혹은 분비에 있어서 중요한 역할을 하는 유전자라 추측이 된다.
