ERC (ELKS-Rab6-interaction protein-CAST) is a family of multidomain proteins with two known members (ERC1 and ERC2). ERC is implicated in the organization of presynaptic active zones, but the underlying mechanism is poorly understood. In the first part of this study, I demonstrate that ERC directly interacts with liprin- α/SYD2, a family of multidomain proteins involved in the development of presynaptic active zones. In vitro results including yeast two hybrid, pulldown, coimmunoprecipitation and coclustering assays indicate that ERC2, an active-zone specific isoform, interacts with all of the known four isoforms of liprin-α and that liprin-α1 associates with both ERC2 and ERC1b, a splice variant of ERC1 that distributes to both cytosolic and active zone regions. ERC2 colocalizes with liprin-α1 in cultured neurons and forms a complex with liprin-α1 in rat brain. liprin-α1, when expressed alone in cultured neurons, shows a partial synaptic localization. When coexpressed with ERC2, however, liprin-α1 is redistributed to synaptic sites. Moreover, roughly the first half of ERC2, which contains the liprin-α-binding region, is sufficient for the synaptic loalization of liprin-α1 while the second half is not. In the second part, I identify that ERC directly interacts with syntenin-1, a tandem PDZ domain-containing protein. In vitro experiments indicate that syntenin-1 interacts with ERC1b and ERC2. mRNAs of syntenin-1 and ERC1b/2 show overlapping expression patterns in rat brains. Syntenin-1 and ERC proteins partially colocalize in dissociated cultured hippocampal neurons, and form a complex in rat brain. Interestingly, expression of active-zone specific ERC2 in cultured neurons induces a marked presynaptic clustering of both endogenous and exogenous syntenin-1. This clustering requires, in addition to the PDZ interaction, the multimerization of both proteins. Importantly, neuronal activity selectively increases the expression levels and synaptic localizations of ERC2, but not those of RIM1 and syntenin-1. The increase in synaptic localization of ERC2 occurs in both dendrites and axons, suggesting that synaptic activity may generally regulate the synaptic locales of ERC2 in vivo. Knockdown of syntenin-1 by small interference RNA approach shows selectively decreases the number of ERC2 puncta, indicating that syntenin-1 is required for the maintenance of ERC2 in neurons. These results suggest that ERC2 regulates the presynaptic clustering of syntenin-1 via PDZ interaction and protein multimerization and that ERC2-dependent active zone assembly through syntenin-1 may be regulated by synaptic activity. Taken together, these results suggest that ERC regulates the assembly of presynaptic active zones by its functional interactions with liprin-α and syntenin
ERC 단백질은 ERC1과 ERC2로 구성되어 있는 단백질로서 전시냅스 접합부를 구성하는데 중요하나 그 정확한 기작에 대해서는 알려져 있지 않다. 첫번째 연구는 ERC 단백질과 liprin 단백질의 상호작용과 그 기능에 관한 것이다. yeast two hybrid, pulldown, coimmunoprecipitation, coclustering assay 등의 in vitro 실험들을 통해서 두 단백질의 상호작용을 증명하였다. 신경세포에서 부분적으로 synapse에 위치하는 liprin-alpha 단백질은 ERC와 같이 발현시키면, 시냅스로 이동하게 되며, 이러한 과정에는 ERC 단백질의 N-terminus 부분이 관여함을 밝혔다. 두번째 연구는 ERC 단백질과 syntenin 단백질간의 상호작용과 그 기능에 관한 것이다. in vitro 실험들을 통해서 두 단백질간의 상호작용을 증명하였고, in situ hybridization을 통해서 mRNA 발현과 단백질 발현 수준에서 두 단백질간의 공통적인 특질들을 발견하였다. 신경세포에서 liprin-alpha와 마찬가지로 syntenin도 부분적으로 시냅스에 위치하게 되는데, ERC를 같이 발현시키면, 역시 마찬가지로 시냅스로 이동하게 된다. 이러한 과정에는 두 단백질간의 직접적인 상호작용 이외에 multimerization 기작이 작용함을 밝혔다. 흥미롭게도 시냅스 활성에 따라서 선택적으로 ERC2의 발현 수준이 증가하게 되고, 이는 전시냅스 접합부의 기능적인 측면에서 의미가 있음을 제시하였다. 마지막으로, syntenin-1의 발현수준을 RNAi 방법을 이용해서 낮추어주었는데, 이때 선택적으로 ERC2를 포함한 몇몇 단백질들의 시냅스 위치가 변동되었다. 이는 syntenin-1이 ERC2가 시냅스에서 유지되는데 필요하다는 얘기가 된다. 이러한 결과들에 비추어볼 때, ERC 단백질은 liprin-alpha와 syntenin-1과 함께 전시냅스의 기능적 유지에 필요하다고 결론지을 수 있다.
