Metastasis is main cause of death in cancer patients. Active matrix metalloproteinases (MMPs) play a critical role in tumor invasion and metastasis. Many studies have shown that reactive oxygen species (ROS) increase the expression level and activation of MMPs through mitogen-activating protein kinase (MAPK) signaling pathway. In a cell, hydrogen peroxide, a kind of ROS, is metabolized hydroxyl radical by intracellular iron ions. Treatment of iron chelator desferrioxamine (DFO) co-treated with PMS for sustained hydrogen peroxide generation accumulates intracellular hydrogen peroxide that induces the up-regulation of MMP-9. The MMP-9 expression is activated through p38 MAPK signaling pathway. The phosphorylation of p38 MAPK is increased as the intracellular hydrogen peroxide accumulated. Phosphorylated p38 MAPK increases the translocation of NF-κB into a nucleus. Inhibition of p38 MAPK signaling pathway decreases the NF-κB translocation. And inhibition of NF-κB activation reduces the MMP-9 expression level. So, it is found that DFO-PMS co-treatment accumulates intracellular hydrogen peroxide that activate the MMP-9 expression level through p38/NF-κB pathway.
암 전이는 암에 의한 사망의 주요한 원인이다. 이러한 암의 전이 과정에서 세포 기질을 분해하는 MMP의 역할이 중요한 것으로 알려져 있다. 따라서 MMP 유전자의 발현 조절과 활성화에 의해 암의 전이정도가 달라지게 된다. 최근 여러 연구를 통하여 세포 내의 활성 산소종, 특히 과산화 수소가 이러한 MMP 유전자 발현을 증가시키는 것으로 알려져 있다. 특히 몇몇 암세포들의 경우 정상 세포에서보다 세포 내의 활성 산소종의 양이 높게 나타나는 것으로 알려져 있다. 이러한 활성 산소종은 세포내 단백질, 지질, 핵산을 변형시키기 때문에 세포 내에서 효소에 의해서 혹은 철 이온에 의해 대사되어 제거된다.
본 연구에서는 세포내 활성 산소종이 증가한 암세포에서 철 킬레이터인 DFO가 MMP-9의 발현에 어떠한 영향을 미치는지 살펴보았다. 인간 유방암 세포인 MCF-7에서 DFO와 PMS를 동시에 처리하였을 때 발생하는 과산화수소의 양은 각각을 처리하였을 때보다 증가하였으며 이에 따라서 MMP-9의 발현양이 증가하였다. 또한 이 때의 MMP-9의 발현 증가는 p38 MAPK에 의해 NF-kB가 활성화 되어 유전자 발현이 조절되는 것임을 확인하였다.
최근 항암 치료제로 철 킬레이터가 많이 연구되고 있다. 철은 세포 성장에 반드시 필요한 원소이기 때문에 철 킬레이터에 의해서 암세포의 성장이 억제된다. 그러나 철 이온에 의한 과산화수소의 대사과정이 차단되기 때문에 MMP 유전자의 발현을 증가시켜 암 전이를 촉진할 수 있음을 확인하였다. 따라서 항암 치료에 사용하는 철 킬레이터들에 대한 더 많은 연구가 필요하다.
