The proteins interacting with speckled POZ domain protein SPOP were sought to clarify the biophysical roles of SPOP. SPOP was first identified as an auto-antigen from the serum of a scleroderma patient and known to interact with Daxx. The interaction between Daxx and SPOP are reportedly known to modulate Daxx-mediated transcriptional regulation. Steroid receptor coactivator-1 (SRC-1), a known coactivator of estrogen receptor (ER), was found to interact with SPOP in the yeast two-hybrid system. This study presents the biochemical and functional interactions between SPOP and SRC-1. In vitro binding and GST pull-down test proposed a direct interaction between SPOP and SRC-1. The N-terminal region of SRC-1 interacts with the N-terminal and C-terminal regions of SPOP. Data further demonstrate that SPOP represses ER-mediated transcription and reverses the activation of ER-mediated transcription by SRC-1. This study proposes that SPOP plays an essential role in modulating SRC-1-mediated transcriptional activation of ER activity.
SPOP의 생리적 기능을 알아보기 위해 SPOP과 결합하는 단백질을 찾아보았다. SPOP은 경화증 환자의 자가항체에 의해 인식되는 새로운 항원으로 발견되었고, Daxx 단백질과 결합하는 것으로 알려졌다. SPOP은 Daxx와 결합하여 Daxx에 의한 전사 조절에 관여한다. 효모를 이용한 결합 단백질 선별 검사를 통해 에스트로겐 수용체의 활성 보체로 알려진 steroid receptor coactivator-1 (SRC-1)이 SPOP과 결합함을 알았다. 세포 외에서의 단백질 결합과 GST pull-down 실험에서 SPOP과 SRC-1이 직접적으로 결합하였다. 두 단백질의 결합에는 SRC-1의 N-말단과 SPOP의 N-말단, C-말단이 중요하였다. 그리고 SPOP은 SRC-1에 의해 증가된 에스트로겐 수용체의 전사 활성을 억제하였다. 따라서 SPOP이 SRC-1이 조절하는 에스트로겐 수용체의 전사적 활성을 보다 정밀하게 조절하는 인자라는 추측이 가능하다.
