Metabolic engineering studies for the production of various metabolites in Escherichia coli have been carried out. For these tasks, metabolic flux analysis (MFA) technique was applied to identify metabolic condition of recombinant E. coli strains.
At first, a pfl ldhA double mutant E. coli strain NZN111 was used to produce succinic acid by overexpressing the E. coli malic enzyme. To have an insight into the intracellular metabolism, MFA was carried out. From the result, it was predicted that supplying additional reducing power could enhance succinic acid production, and more reduced carbon substrate sorbitol was thus examined for the possibility of matching the potential during succinic acid production. When NZN111 (pTrcML) was cultured in LB medium containing 20 g/L sorbitol under CO2 atmosphere, 10 g/L of succinic acid was produced, and the apparent yield of succinic acid was 1.1 g succinic acid/g sorbitol.
Secondly, the metabolic network of E. coli was constructed, and was used to simulate the distribution of metabolic fluxes in recombinant E. coli producing poly(3-hydroxybutyrate) [P(3HB)]. It was notable to find from MFA result that the Entner-Doudoroff (ED) pathway flux increased significantly under P(3HB) accumulating condition. The P(3HB) content obtained with KEDA (pJC4), which is defective in the activity of Eda, was lower than that obtained with its parent strain. The reduced P(3HB) biosynthetic capacity of KEDA (pJC4) could be restored by the coexpression of the E. coli eda gene, which proves the important role of ED pathway on P(3HB) synthesis in recombinant E. coli as predicted by metabolic flux analysis.
Thirdly, a new strategy, metabolic flux profiling and clustering, was proposed and applied to systematically study the variation of metabolic fluxes in E. coli under metabolite production conditions. Through those analyses, I was able to predict the real achievable maximum yields of metabolites rather than theoretical maximum yields, which can be used as guidelines on metabolic engineering studies. I also applied those strategies for the identification of core fluxes controlling metabolic activity, and it was proven that enhanced metabolites production could be achieved by modification of core fluxes.
대사흐름분석기술(metabolic flux analysis)을 이용하여 다양한 대사산물을 생산하는 대장균의 대사특성을 분석하고, 이를 바탕으로 균주의 대사특성을 개량하였다.
첫째로, 숙신산을 생산하는 대장균의 대사특성분석을 수행하였다. 본 연구를 통하여 pyruvate를 거쳐서 숙신산을 생산하는 새로운 숙신산 생산 pathway를 제안하였으며, 이 경우 말릭산을 숙신산으로 변환하는 pathway가 bottleneck임을 확인할 수 있었다. 대사조적분석(metabolic control analysis)결과 환원력(reducing power)이 숙신산 생산에 중요한 역할을 한다는 사실을 확인할 수 있었다. 부족한 환원력을 공급하기 위하여 sorbitol을 탄소원으로 사용한 결과 1.1 g succinic acid/g glucose의 높은 숙신산의 수율을 얻을 수 있었다.
둘째로, poly(3-hydroxybutyric acid) [P(3HB)]를 생산하는 대장균의 대사흐름을 분석하였다. 전체 건체중량의 49% 와 78%의 P(3HB)를 생산하는 대장균의 대사흐름 변화를 분석한 결과, 대장균내에서 활성화되지 않는다고 알려진 Entner-Doudoroff (ED) pathway가 활성화 됨을 확인할 수 있었다. ED pathway의 중요성을 실험적으로 검증하기 위하여 ED pathway상의 eda 유전자 돌연변이 균주를 제작하여, P(3HB)생산 실험을 수행하였다. 실험결과 eda 유전자 돌연변이의 P(3HB)생산능이 감소함을 확인하였으며, eda 유전자 돌연변이 균주에 eda 유전자를 재도입할 경우 P(3HB)생산량이 회복됨을 확인하였다.
마지막으로, 균주의 대사특성을 파악하기 위하여 metabolic flux profiling과 metabolic flux clustering이라는 새로운 전략을 제안하였다. Metabolic flux profiling기법을 초산, 유산, P(3HB), leptin을 생산하는 재조합 대장균에 적용하였으며, 이를 통하여 이들 대사산물의 실제 생산가능한 최대수율을 예측할 수 있었다. 또한, metabolic flux clustering기법을 적용함으로써, 초산, 유산, P(3HB)의 생산에 해당회로가 중요한 역할을 함을 예측하였다. 이의 검증을 위하여 해당회로상의 tpiA 유전자를 도입하였으며, 이 경우 초산 및 P(3HB)의 생산량이 증가함을 확인할 수 있었다.
