Kaposi’s sarcoma (KS)-associated herpesvirus (KSHV) is the eighth type of human herpesvirus (HHV8) and human gamma herpesvirus related to Epstein-Barr virus (EBV). KSHV has been implicated in the pathogenesis of Kaposi’s sarcoma and it encodes putative oncogenes and genes that may cause Kaposi’s sarcoma pathogenesis. KSHV open reading frame K8 encodes a protein of 237 amino acids with a prototypic basic region-leucine zipper (bZIP) domain at the carboxyl-terminus by in-frame splicing. KSHV K8 shows significant homology to the EBV immediate-early protein Zta, a key regulator in the reactivation and replication of EBV. In this study, we report that K8, like its homolog EBV Zta, interacts with cellular CREB-binding protein (CBP) in vivo and in vitro. This interaction requires the C/H3 domain of CBP and the basic region of K8. K8 represses CBP-mediated transcription by competing limited amounts of cellular CBP, exemplified by the reduced expression from the AP-1 and human immunodeficiency virus long terminal repeat promoters.
사람에 감염되어 숙주세포 내에서 잠복기를 거치는 카포시 육종 허피스 바이러스는 감마 허피스 바이러스로서 8번째로 발견된 인간에 감염되는 허피스 바이러스이다. 카포시 육종 허피스 바이러스open reading frame K8은 237개의 아미노산으로 이루어져 있으며 basic region-leucine zipper domain을 이용하여 homodimerization을 하는 단백질을 만들어 낸다. K8 단백질은 같은 gammaherpesvirinae에 속하며 동일한 잠복기의 특성을 가지는 Epstein Barr virus (EBV)의 reactivation과 replication 의 핵심 조절자인 immediate-early protein Zta 와 아주 큰 유사성을 보인다. 본 연구에서는 K8이 유사체인 EBV Zta처럼 세포 내 CREB-binding protein (CBP)과 생체조건 안과 밖에서 모두 결합하며, 이러한 상호작용을 위해서는 CBP의 C/H3 domain과 K8의 basic region을 필요로 한다는 것을 밝혀 내었다. K8은 AP-1과 human immunodeficiency virus long terminal repeat의 promoter로부터의 발현을 억제하는 실험결과서 알 수 있듯이, 이러한 상호작용을 통해 세포 내에 소량으로 존재하는 CBP를 다른 단백질들과 경쟁함으로써 CBP에 의해 중재되는 전사를 억제한다.
