Sodium butyrate (NaBu) can enhance the expression of genes from some of the mammalian promoters including the CMV and SV40 while it can also inhibit cell growth and induce cellular apoptosis. Thus, the beneficial effect of using a higher concentration of NaBu on a foreign protein expression is compromised by its cytotoxic effect on cell growth. To overcome this cytotoxic effect of NaBu, a survival protein, human Bcl-2, was overexpressed in the recombinant CHO cells (SH2-0.32) producing a humanized antibody directed against the S surface antigen of hepatitis B virus. When batch cultures of both control cells transfected with bcl-2 deficient plasmid (SH2-0.32-Δbcl-2) and cells transfected with bcl-2 expression plasmid (14C6-bcl-2) were performed in the absence of NaBu, both cells showed similar profiles of cell viability and antibody production. Compared with SH2-0.32-Δbcl-2 culture, under the condition of NaBu addition at the exponential growth phase, overexpression of bcl-2 gene considerably suppressed the NaBu-induced apoptosis of 14C6-bcl-2 by inhibiting caspase-3 activity and extending the culture longevity by more than 2 days. As a result, the final antibody concentration of 14C6-bcl-2 culture was 2 times higher than that of SH2-0.32-Δbcl-2 culture in the presence of NaBu and 3 times higher than that of SH2-0.32-Δbcl-2 and 14C6-bcl-2 cultures in the absence of NaBu. Overexpression of human Bcl-2 protein in SH2-0.32 considerably suppressed NaBu-induced apoptosis during batch culture using a commercially available serum-free medium, extending the culture longevity. Due to the enhanced transcription efficiency and the extended culture longevity, the final antibody concentration of 14C6-bcl-2 culture (23㎍/mL) was 2 times higher than that of SH2-0.32-Δbcl-2 culture (10.5㎍/mL) in the presence of NaBu. To determine the effect of NaBu/Bcl-2 overexpression on the molecular integrity of protein products, antibodies purified from 14C6-bcl-2 and SH2-0.32-Δbcl-2 cultures in the presence of NaBu were characterized using various molecular assay systems. No significant changes in molecular weight of antibodies could be observed at the level of SDS-PAGE. From GlycoSep-N column analysis, it was found that the core oligosaccharide structure $(GlcNAc_2Man_3GlcNAc_2)$ was not affected by NaBu/Bcl-2 overexpression while the microheterogeneity of N-linked oligosaccharide structure was slightly affected. Compared with the antibody produced in the absence of NaBu, the proportion of neutral oligosaccharides was increased by 10% (14C6-bcl-2) to 16% (SH2-0.32-Δbcl-2) in the presence of NaBu, which was accompanied by the reduced proportion of acidic oligosaccharides, especially of monosialylated and disialylated forms. The changes in microheterogeneous oligoformal structures of antibody in turn affected the mobility of antibody isoforms in isoelectric focusing (IEF), resulting in the occurrence of some more basic antibody isoforms produced in the presence of NaBu. However, the antigen-antibody binding properties were not changed by alteration in glycosylation pattern. The competitive enzyme-linked immunosorbent assay (ELISA) showed that the antibody produced by NaBu/Bcl-2 overexpression maintained its antigen-antibody binding properties with binding affinity of about $2.5×10^9M^{-1}$. Taken together, no significant effects of NaBu/Bcl-2 overexpression on the molecular integrity of antibodies produced using serum-free medium could be observed by the molecular assay systems.
Apart from the overexpresson of Bcl-2 survival protein to overcome cytotoxic effect of NaBu, the expression vector of antisense RNA of caspase-3 was constructed and transfected to SH2-0.32. Using this antisense RNA strategy, rCHO mutant cells (B3 clone) producing a low-level of caspase-3 proenzyme were isolated. Compared with control cell culture, under the condition of 5 mM NaBu addition at the exponential growth phase, expression of antisense RNA of caspase-3 significantly suppressed the NaBu-induced apoptosis of B3 by more than 2 days. However, compared with control cell culture, the final antibody concentration of B3 culture was not increased in the presence of NaBu, which may be due to the loss of cellular metabolic capability resulted from the depolarization of mitochondrial membrane. Taken together, this study suggests that, although expression of antisense RNA of caspase-3 does not improve antibody productivity of rCHO cells, it can suppress NaBu-induced apoptotic cell death of rCHO cells and thereby may reduce problems associated with cellular disintegration.
The feasibility of Bcl-2 overexpression in improving cell viability and protein productivity under hyperosmolar condition was also investigated by comparing SH2-0.32-Δbcl-2 and 14C6-bcl-2. Various hyperosmolar stresses (294-714 mOsm/kg) were induced by addition of corresponding amounts of NaCl into culture medium. The inhibitory effect of hyperosmolality on cell growth was dose-dependent and the specific antibody productivity $(q_{Ab})$ was proportionally enhanced with increasing medium osmolality. Compared with physiological osmolality (294 mOsm/kg), 2.1- and 2.5-fold enhanced final antibody production could be obtained in SH2-0.32-Δbcl-2 and 14C6-bcl-2 cell cultures, respectively, as medium osmolality increased up to 522 mOsm/kg. The increased $q_Ab$ could be visualized as a shift in intracellular antibody contents analyzed by FACS. However, over 580 mOsm/kg, the final antibody production was not improved despite the elevated $q_Ab$ because the cytotoxic effect of hyperosmolar stress was so significant that a great extent of cell death was induced. The apoptotic cell death began to appear at 522 mOsm/kg in control cells 24 hr after the introduction of hyperosmolar stress. Bcl-2 overexpression could delay the apoptosis induced by 522 mOsm/kg in 14C6-bcl-2 cells by inhibiting caspase-3 activation. Necrotic cell death overwhelmed the apoptosis in both control and 14C-bcl-2 cells over 580 mOsm/kg, where Bcl-2 overexpression failed to improve cell viability. In the range of 294-522 mOsm/kg, the improved cell viability and cell membrane integrity determined by measuring extracellular LDH activity in 14C6-bcl-2 cells can be a good indicator of product integrity produced in a high cell density cultivation and an elevated hyperosmolar culture condition.
뷰티르산염 (sodium butyrate)은 싸이토메갈로 바이러스 (CMV)나 씨미언 바이러스 40 (SV40) 같은 포유동물세포 프로모터로 유도되는 유전자의 발현을 증가시키는 동시에 세포성장을 억제하고 세포예정사 (apoptosis)을 유도할 수 있다. 뷰티르산염의 세포독성을 극복하기 위해 B형 간염 바이러스 (HBV)의 S 표면항원을 생산하는 재조합 Chinese hamster ovary (rCHO) 세포 (SH2-0.32)에서 인간 Bcl-2 생존단백질을 과발현시켰다. 대조군세포 (SH2-0.32-Δbcl-2)와 Bcl-2를 과발현하는 14C6-bcl-2 세포는 뷰티르산염을 첨가하지 않은 회분배양에서 비슷한 세포생존률과 항체생산성을 보였다. 그러나 뷰티르산염을 대수성장기에 첨가한 경우 대조군세포와 비교하였을 때 14C6-bcl-2 는 Bcl-2 단백질의 과발현에 의해 caspase-3 활성화를 억제함으로써 뷰티르산염에 의한 세포예정사 유도를 지연시켜 배양 기간을 2일 이상 증가시킬 수 있었다. 결과적으로 뷰티르산염이 존재하는 경우 회분배양에서 14C6-bcl-2의 최종항체생산성은 대조군세포보다 2배 이상, 뷰티르산염이 존재하지 않는 경우보다 3배 이상 증가하였다.
Bcl-2과발현에 의해 뷰티르산염으로 유도되는 세포예정사의 억제는 무혈청 상용배지에서도 관찰되었는데 뷰티르산염에 의한 전사속도의 증가 및 배양기간의 연장 효과에 의해 14C6-bcl-2의 최종항체생산성은 SH2-0.32-Δbcl-2 세포보다 2배 이상 증가하였다. 뷰티르산염/Bcl-2 과발현 시스템이 생산되는 단백질의 분자적 구조에 어떠한 영향을 줄 수 있는가를 알아보기 위해서 뷰티르산염이 존재하는 무혈청배지에서14C6-bcl-2과 SH2-0.32-Δbcl-2 세포를 배양하여 항체를 분리하였다. 대조군항체로는 뷰티르산염이 없는 상태에서 SH2-0.32 세포로부터 생산된 항체를 사용하였다. 분석결과 SDS-PAGE 상에서 뷰티르산염/Bcl-2 과발현에 의한 항체분자량의 변화는 관찰할 수 없었으며, GlycoSep-N 컬럼 분석에서도 비록 N-연결당쇄구조의 미세구조에는 약간의 변화가 있지만 핵당쇄구조 $(GlcNAc_2Man_3GlcNAc_2$)의 변화는 관찰되지 않았다. 대조군항체와 비교시 중성당쇄구조의 비율이14C6-bcl-2과 SH2-0.32-Δbcl-2 세포에서 각각 10%와 16%씩 증가하였으며 이와 동시에 말단 시알산 (sialic acid) 이 한 개, 혹은 두 개가 연결된 산성당쇄구조의 비율은 약간 감소하였다. 이러한 당쇄의 미세구조 변화는 IEF 분석에서 항체의 염기성 이성체의 출현과 관계가 있을 것으로 추정되었다. 당쇄의 미세구조 변화에도 불구하고 뷰티르산염/Bcl-2 발현 시스템에 의해 생성된 항체는 대조군항체와 동일한 항원-항체의 결합성을 유지하고 있었다. 결과적으로 무혈청배지에서 뷰티르산염/Bcl-2 발현시스템에 의한 항체의 분자적 구조에는 커다란 영향을 주지 않았음을 알 수 있었다.
뷰티르산염에 의한 세포독성을 극복하기 위해 Bcl-2 생존단백질 과발현과는 별도로 SH2-0.32 세포에 caspase-3에 대한 antisense RNA를 발현하는 벡터를 도입하였고 caspase-3 proenzyme의 발현이 억제된B3 clone을 얻을 수 있었다. 대수성장기에 5 mM의 뷰티르산염을 첨가하였을 때 B3 세포에서는 뷰티르산염에 의한 세포예정사를 억제하여 대조군세포 (SH2-0.32-ΔASCASP3-200)와 비교하여 2일 이상 배양기간의 연장을 관찰할 수 있었다. 그러나 뷰티르산염이 존재하는 환경에서 대조군세포와 비교하여 B3 세포의 최종항체생산성은 증가하지 않았는데 이것은 뷰티르산염에 의해 미토콘드리아의 막전위가 탈분극되어 세포의 대사능력이 떨어지기 때문인 것으로 추정되었다. 비록 antisense RNA를 이용하여 뷰티르산염에 의한 항체생산성을 증가시킬 수는 없었지만 세포예정사를 억제, 세포생존성을 증가시킴으로써 죽은 세포의 분해로 인한 단백질 생산성 및 안정성의 문제를 어느 정도 감소시킬 수 있을 것으로 여겨진다.
14C6-bcl-2과 SH2-0.32-Δbcl-2 대조군세포를 다양한 고삼투압 배지에서 배양하여 Bcl-2 생존단백질이 고삼투압 환경에서 세포생존률과 항체생산성을 향상시킬 수 있는가를 알아보았다. 고삼투압에 의한 세포성장 억제 효과는 삼투압에 비례하여 나타났으며 비항체생산성 $(q_Ab)$ 은 삼투압이 증가함에 따라서 비례적으로 증가하였다. 정상삼투압 (294 mOsm/kg)과 비교하여, 522 mOsm/kg까지 삼투압이 증가함에 따라 최종항체생산성은 대조군세포와14C6-bcl-2에서 각각 2.1배와 2.5 배 증가하였다. 고삼투압에 의한 $q_Ab$의 증가는 FACS 분석에서 세포내 항체함량의 증가로도 관찰할 수 있었다. 580 mOsm/kg이상의 삼투압에서는 비록 $q_{Ab}$ 는 증가하였지만 고삼투압에 의한 세포독성에 의해 과도한 세포사가 유도되어 최종항체생선성은 증가하지 않았다. 고삼투압 세포독성에 의한 세포예정사는 대조군세포에서 522 mOsm/kg 삼투압을 처리한 24시간 이후부터 나타나기 시작했으며 14C6-bcl-2세포에서의 Bcl-2 과발현은 caspase-3의 활성화를 억제함으로써 522 mOsm/kg 삼투압에 의한 세포예정사의 유도를 억제할 수 있었다. 그러나 580 mOsm/kg이상의 고삼투압에서는 세포예정사보다는 세포괴사 (necrosis)가 주로 관찰되었으며 이러한 세포괴사는 Bcl-2 과발현에 의해서 억제되지 않았다. 294-522 mOsm/kg 삼투압 범위 내에서 14C6-bcl-2의 Bcl-2 과발현에 의한 세포생존률 및 세포막의 안정성 향상은 배지내 LDH 효소 활성의 감소로 확인할 수 있었으며 이러한 결과는 결국 고농도세포배양 및 고삼투압 환경에서 단백질의 안정성 유지에 유리할 수 있음을 말해 주고 있다.
