Bdellin is a potent trypsin, plamin and acrosin inhibitor purified from the leech. It is expressed in all parts of the body with high concentration in the reproductive organs.Bdellin-KL, bdellin from the Korean leech, has unsual hisitidine reach repeats which may be related to its function.But the physiological function in the leech has not revealed yet. To study on bdellin in gene level,the genomic clone encoding bdellin-KL was isolated from the constructed leech library.A positive clone containing the ORF which consists of 3 exons and 2 introns spanning about 1kb and 5'- and 3'- flanking region was isolated.The nucleotide sequences of exon-intron junctions were in agreement with the consensus 5'-GT …AG- 3'.To characterize the promoter, a transcription start point (tsp) was identified and the 5'-flanking sequence was analyzed.The consensus TATA box sequence (TATAAA) was found at - 28 bp from the tsp. Two CAAT boxes and two GC boxes were located upstream of the TATA box.Unusual ATA repeats were found upstream of the detected transcription factor binding sites.The promoter contained several putative cis-elements, which may serve as the binding sites of the important transcription factors related to the physiological function of bdellin.
Bdellin - KL은 Hirudo nipponia로부터 분리된 bdellin으로서 trypsin, plasmin, acrosin inhibition activity를 가지고 있다. Bdellin의 생리적 기능을 알기 위한 연구의 하나로서 bdellin-KL의 유전자를 library의 screening을 통해 클로닝하고 그 염기서열을 분석하였다.
전체 open reading frame과 5', 3'인접 서열들을 가지고 있는 clone을 찾아낸 후 그 구조를 분석한 결과 이 유전자는 3개의 exon과 2개의 intron으로 이루어졌으며 splicing position에서 GT-AG rule을 따르고 있음이 밝혀졌다. Primer extension analysis 를 통해 transcription 시작점이 시작 codon으로부터 약 60 염기쌍 정도 떨어진 곳임을 알 수 있었고 그 곳에서 28염기쌍 위 쪽으로 TATA box가 존재하고 있음을 밝혔다. 각각 두 곳의 GC, CAAT box 서열이 TATA box 위 쪽으로 위치해 있었고 AP-1, AP-3 결합 부위가 있었다. 이러한 factor들의 전체 혹은 일부가 bdellin의 조직 비특이적 발현을 조절하고 있을 것으로 생각되었다. 조직 특이성을 가지고 발현되는 몇 가지의 transcription factor들이 binding할 수 있는 서열들을 찾아내었는데 대표적으로 GATA-1, NF-E, E2A 결합 부위, E-box 들을 포함하고 있었다. 이 factor들은 bdellin의 거머리 내에서의 생리적 기능과 밀접한 연관을 맺을 수 있다고 생각되었으며 이들이 어떻게 bdellin-KL의 발현을 조절하고 있는지를 연구할 필요성이 있음을 알 수 있었다.
