Two cDNAs encoding pyruvate dehydrogenas(PD) and citrate synthase(CS) related to citrate were isolated and characterized from tomato plants. Two clones PD, CS were 1416 bp and 1176 bp, respectively and had a open reading frames(ORF), deriving 52kDa and 43kDap olypeptide. The identities between the deduced amino acid sequences of tomato CS and potato CS were 98%. For PD of tomato and potato same identities (98%) were shown.
Genomic DNA blot analysis suggests that MD,PD and CS may be single copy genes. The high est expression of PD, CS and MD (malate dehydrogenase) occurred in roots and MD, PD and CS were expressed in fruit at all developmental stages, and their expression levels were relatively similar. The present study suggests that the expression of TCA cycle was not uniform in the different tissues of the tomato. Although mitochondrial citrate synthase and pyruvate dehydrogenase are usually described as a housekeeping enzyme, its expression is developmentally regulated, indicating that in certain tissues or under certain environmental conditions a higher amount of the enzyme is required.
토마토로부터 시트르산에 관계된 효소인 pyruvate dehydrogenase 와 citrate synthase를 분리하였고 분석하였다. PD, CS로 명명된 두유전자는 각각 1176, 1416개의 염기쌍 으로 구성되어 있었고, 각 유전자는 43kDa과 52kDa 크기의 폴리펩티드를 암호화하는 하나의 open reading frame(ORF)를 구성하고 있었다.
분리된 토마토의 PD와 감자의 PD유전자와의 아미노산서열의 상동성은 98%로 나타났고 토마토와 감자의 CS 유전자 역시 98%의 상동성을 나타내었다. 토마토 게놈에 대한 서던 분석결과로, MD,PD와 CS는 게놈내에 단일 카피로 존재할 것이라고 추정된다.
MD, PD 그리고 CS는 토마토 뿌리에서 가장 강하게 발현되었고 잎과 줄기 과육에서는 상대적으로 약하게 발현되었으나 모두 명백한 발현이 관찰되었다. MD, PD 그리고 CS는 모든 과육 발달단계에서 발현되었고 그 발현정도는 과육 발달단계에 관계없이 거의 비슷했다. 결과적으로 시트르산 생성에 관여하는 세 유전자는 어느 조직에서나 어느 발달단계에서나 지속적인 발현이 있고 그 발현 정도도 비교적 높은 편이다.
