Human papillomavirus is the causative agent of cervical cancer. Using genetic and biochemical approaches, we provide the evidence that HPV E7 is able to associate with cAMP response element binding protein binding protein (CBP) and the related protein p300 in vivo and in vitro. Transient transfection assays showed that HPV E7 enhances the transcriptional activation functions of CBP. These results suggest that E7 can stimulate some cellular CBP mediated promoters. In addition, we show that HPV E7 binds to HPV E6 in vitro, and E7 also suppresses IRF-1 dependent promoter. From these data, we speculate that both E6 and E7 disturb the cellular interferon pathway, and CBP is a mediator of these interference activities of HPV E6 and E7.
파필로마바이러스는 자궁 경부암의 원인으로 알려져 있다. 유전학적, 생화학적 방법을 이용하여 파필로마바이러스 E7 단백질이 CBP와 시험관 내, 외에서 결합함을 알아내었다. 또한 파필로마바이러스 E7은 CBP의 전사촉진 기능을 향상시킬 수 있음을 알 수 있었다. 이러한 결과는 E7이 CBP에 의해 영향을 받는 프로모터에 관여할 수 있음을 제시한다. 게다가 E7은 E6와 시험관 내에서 결합함을 알 수 있었으며 E7은 또한 IRF-1 및 IRF-3와 결합함을 알 수 있었다. IRF-1이 E7에 의해 저해되는 현상으로부터 미루어 보아 E6, E7은 세포 내 인터페론 기작을 저해하며, 나아가 CBP를 이러한 작용의 매개로 사용함을 유추할 수 있었다.
