The aim of this study is to examine the brain dynamics in patients with Alzheimer''s disease by the simulation of the EEG in the progress of Alzheimer''s disease using neural network constructed based on biological anatomic structure of cerebral cortex, the mechanism of EEG generation and the Freeman''s asymmetry sigmoid nonlinearity. The simulated EEG have the comparable dynamical properties in accord with physiological findings of the human EEG. The spectrum of the simulated EEG is similar to that of the real EEG seen in the alert state with components in the delta, theta, alpha, beta ranges. The simulated EEG has the similar values of the correlation dimension (D2), the first positive Lyapunov exponent (L1), and Kolmogorov-Sinai entropy (KS-entropy), which are measures of complexity. These nonlinear estimates revealed that the simulated EEG is a complex and chaotic time series. Determinism test showed that it does not have low-dimensional determinism but nonlinearity as real EEG does. We investigate the changes of linear and nonlinear properties of the simulated EEG as the synaptic densities of the neural network decrease like the cerebral cortex of the brain damaged by Alzheimer''s disease. We show that the mean frequency of the power spectrum of the simulated EEG shifts to low frequencies as the synaptic densities decrease. The values of the D2, the L1, and the KS-entropy also decrease as the network deteriorates. These results are comparable nature of the EEG in patients with Alzheimer''s disease. Especially the decreases of synaptic densities of long corticocortical connection (alpha connection) with fixed subcortical connections (beta connections) lead to the power spectrum of the simulated EEG shift to low frequencies and lowered complexities estimated by the D2, the L1 and the KS entropy, while the decreases of subcortical connections with fixed long corticocortical connections result in no significant changes in those nonlinear measures. This result may support the hypothesis that Alzheimer''s disease is a neocortical disconnection syndrome in which there is a loss of structural and functional integrity of long corticocortical tracts. In this study we propose that EEG simulation by computational neural network can be a useful tool in understanding the brain dynamics in the progress of Alzheimer''s disease or other brain diseases.'
본 연구의 목적은 실제 사람 뇌의 해부학적 구조와 신경세포군의 비선형적 특성을 고려하여 설계된 신경망을 이용해 알츠하이머 치매 환자의 뇌파를 모델링하고 만들어진 뇌파의 동역학적 성질을 조사해 봄으로써 알츠하이머 치매의 원인을 물리학적으로 기술하는데 있다. WJ 프리만과 JJ 라이트가 각각 독립적으로 제안한 뇌파 발생 신경망을 새롭게 변형한 후 뇌파를 발생할 수 있는 신경망을 구축하였다. 이때 신경망에 의해 발생된 뇌파는 실제 뇌파의 역학학적 성질과 유사한 동역학적 특성을 가지고 있었다. 신경망에서 만들어진 뇌파의 파워 스펙트럼는 실제 뇌파와 유사한 주파수 영역을 가지고 있었으며, 실제 뇌파와 마찬가지로 알파파, 베타파, 델타파, 세타파 등으로 나눌 수 있었다. 비선형 분석을 통해 측정한 상관 차원, 리아프노프 지수, KS-엔트로피 값도 실제 뇌파에서 얻은 값들과 유사했다. 신경망에 의해 발생된 뇌파 역시 상당히 복잡하고 카오스적인 신호라는 사실도 알아내었다. 우리는 알츠하이머 치매의 원인인 신경 세포 수축, 세포 수 감소와 같은 상황을 만들어주기 위하여 신경망의 시냅스 연결을 차례로 끊어가면서 뇌파의 동역학적·카오스적 성질이 어떻게 바뀌는가에 대해 알아보았다. 그 결과 신경망의 시냅스 연결이 끊어질수록 파워 스펙트럼의 주파수가 낮은 주파수 영역으로 이동하고, 상관 차원, 리아프노프 지수, KS-엔트로피의 값이 점점 줄어드는 것을 발견할 수 있었다. 이것은 실제 알츠하이머 환자의 뇌파에서 일어나는 변화와 유사한 상황이었고, 알츠하이머 치매 환자 뇌파의 동역학적 변화의 원인이 ''시냅스 연결의 결핍''에 의한 것이라는 사실을 뒷받침해 주고 있는 것이다. 그 중에서도 작은 컬럼 안의 짧은 연결(베타 연결)보다는 서로 다른 영역을 연결해주는 콜린계 긴 연결(알파 연결)이 더 중요한 역할을 한다는 것을 알 수 있었다. 이는 그 동안 과학자들이 알츠하이머 치매의 원인을 콜린계 긴 연결의 결핍에 의한 연합 기능 약화로 보는 가설을 간접적으로 뒷받침하는 것으로 해석해 볼 수 있다.
