The structure and activity of a helical antimicrobial peptide with a central proline (P14) and its alanine derivative (P14A) were investigated by various spectroscopic methods, restrained molecular dynamics, and biological assay. Both P14 and P14A exhibited cooperative helix formation in TFE solution, but P14 showed much reduced helical stability than P14A. The model structure exhibited a pronounced kink in P14, in contrast to the uniform helix of P14A. The two peptides showed comparable binding to negatively charged lipids, but P14 had considerably reduced affinity to a neutral lipid. With a destabilized helix, P14 exhibited greater antibacterial activity than P14A. It is presumed that the electrostatic interaction between helical peptides and lipid membranes is the dominant factor for antibacterial activity. Moreover, helical stability can modulate the membrane binding driven by electrostatic interaction. The increased antibacterial activity of P14 implies that the helical kink may play an important role in the disruption of bacterial membranes.
Temperature coefficients of amide protons have been investigated by 2D NMR spectroscopy for helical peptides with a central proline (P14) and its alanine derivative (P14A) in an aqueous trifluoroethanol (TFE) solution. The profiles of chemical shifts, exchange rates and solvent perturbations of amide protons are compared with that of temperature coefficients. The periodicity of temperature coefficients coincided exactly with that of chemical shifts. Temperature coefficients of each residue for three different TFE concentrations were obtained for comparison. Upon temperature elevation, the hydrophobic interactions which might be responsible for helical curvature would be weakened. If the helical curvature is diminished during the thermal unfolding process, the most affected residues would be those on the concave (hydrophobic) side of the helix. The unusual behavior of temperature coefficients of amide protons in TFE solution may be partly explained by the variation of hydrogen bond length.
Secondary structure of misgurin, an antimicrobial peptide from the loach is studied by CD and NMR spectroscopy. The peptide was highly hydrophilic and showed low hydrophobic moment. Misgurin had random coil structure in water, and adopted helical structure in TFE solution. The α proton chemical shift exhibited typical upfield shift of a helix, but chemical shift index method implied marginal stability of the helix. The amide proton chemical shift showed no periodicity which is generally observed in amphiphilic helix. Hydrogen exchange study revealed that C terminal moiety was most stable in the helix.
NMR 분광법을 이용하여, 개구리에서 분리된 항균성 펩티드와 그 유도체의 구조를 규명하고 생체막과의 작용을 CD, florescence 분광법을 이용 하여 연구하였다. 이 펩티드는 helix를 이루면서 생체막에 작용하여 항균활성을 보이는데 중앙에 helix를 불안정하게 하는 proline을 가지고 있다. 이 펩티드(P14)와 proline을 alanine으로 치환시킨 유도체(P14A)의 구조를 규명한 결과 P14는 중앙에 kink를 가지며 kink 안쪽이 안정화되는 경향을 보였고 P14A는 정상 helix를 이루는 것을 보였고, 이들 helix의 안정도에 있어서 P14A가 더 뛰어났다. 생체막과의 작용에 있어서, 음성 생체막과는 비슷한 작용을 보였으나 중성 생체막에는 P14의 작용이 많이 감소하였다. 그러나, 항균활성은 P14가 오히려 더 높은 경향을 보였다. 항균 펩티드의 활성에 있어서 정전기적 작용이 중요하게 작용하는 것으로 추론되고, helix의 안정성과 전하가 생체막과의 작용을 조절할수 있는 요인으로 생각 된다. 한편 helix를 불안정하게 하는 proline이 항균 활성을 높인 다는 사실은 항균 작용 기작을 이해하는데와, 새로운 항균물질의 설계에 도움을 주리라 생각된다.
NMR 분광법에서 Amide proton의 온도 상수가 작을 경우, 내부 수소 결합을 알리는 지표로 사용되어 왔다. 그러나 유기 용매를 사용할 경우 맞지 않는 경우가 보고된바 있다. 이 실험에서는 amide proton의 치환 실험과 온도 상수를 비교한 결과 가장 강한 수소결합을 이루는 amide proton이 가장 높은 온도 상수를 보이는 것을 보였다. Solvent perturbation 실험 등으로 이들이 수소결합을 이루는 것을 확인하였다. 이 실험 결과로는 amide proton의 온도 상수는 수소 결합 세기 보다는 그 변화 정도를 알리는 지표로 쓰이는 것이 타당 하다고 생각된다. 짧은 길이의 펩티드들은 TFE등의 유기 용매에서 많이 연구되는데 온도 상수를 수소 결합과 연관짓는 데는 주의가 필요하고 다른 추가 자료가 요구되어야 할 것이다.
미꾸라지에서 분리된 항균성 펩티드인 misgurin의 2차구조를 CD 및 NMR 분광학을 이용하여 연구하였다. Misgurin은 물에서 구조가 없었고 TFE에서 helix 구조를 가지는데, 그 안정도는 크지 않았다. Amide proton 치환 실험에서 C 말단 부위의 helix가 N 말단보다 더 안정된 것으로 나타났다. Misgurin은 적은 양의 helix를 만드는 데에도 불구하고 강한 항균 활성을 보였는데 이는 항균 펩티드의 활성에 정전기적 작용이 크게 작용한다는 사실을 시사 한다. 항균 펩티드의 활성이 helix의 안정도와 반드시 비례하지 않는다는 사실과 함께, 이 결과는 길이를 줄이며 활성을 유지하는 펩티드의 설계에 이용될 수 있을 것이다.
