Strawberry (Fragaria x ananassa Duch.) fruit originates from and is composed of receptacle and achene, which is a unique type differently from the other plant fruits. Although the major part of fruit is fruit-like tissue, it is known that their development is regulated in similar ways to genuine fruit. The complex physiological and biochemical changes in fruit in distinction from other tissues result, at least in part, from changes in the amounts or activities of various enzymes and other proteins present in the fruit tissue.
Four fruit-specific and -dominant proteins (10, 26, 40, 60 KDa proteins) were isolated and determined their N-terminal amino acid sequences. Three proteins with molecular weights of 10, 40, 60 KDa were presumed as proteins with the masked N-teminal. N-terminal amino acid sequence of a protein with molecular weight of 26 KDa showed the significant similarity with plant ascorbate peroxidase (APX) and was found to show APX activity. The results suggested that APX has any function in relation to fruit-specificity.
As the first step to characterize 26KDa protein in molecular level, cDNA encoding the protein was isolated from strawberry fruit. Using degenerate oligonucleotides corresponding to a part of the N-terminal sequence and three sequences corresponding to conserved regions in previously reported plant APX, two RT-PCR products of 730 bp and 630 bp were synthesized. Screening of a strawberry fruit cDNA library with the PCR-amplified DNA fragments as probes resulted in cDNAs that encodes a cytosolic APX (cAPX) isoform. DNA sequenceing revealed that cDNAs contain an open reading frame of 250 amino acids (27.3kDa). The deduced amino acid sequence showed significant homology of about 90% identity with other plant cAPX. The catalytic triad and other functional site were conserved. The lack of a transit peptide in the deduced amino acid sequence implies that it encodes a cytosolic isoform. RNA blot analysis showed that the mRNA is strongly expressed in fruit and weakly in leaf, root, and petiole, but not in seed. Also, during ripening, the expression of the mRNA increased to the maximum level at the turning stage. The isolated twenty-eight clones encoding ApxSC could be grouped into seven clones of APX7, 18, 19, 20, 22, 26, and 27, on the basis of partial sequencing using T7 promoter primer. The results showed that seven groups were different from each other in the nucleotide and amino acid sequence level. The nucleotide sequences had over 90% homology. Seven sequences were closely related to each other, but not identical. In amino acid sequences, three sites were different. The seven sequences indicated expression of at least seven genes. These results are matched with those for the genomic DNA clones. The functional significance, if any, is not known.
To confirm whether isolated ApxSC cDNA is functional and for the other functional studies, an expression plasmid for ApxSC, pETAPX7, has been constructed. It contains a T7 promoter, T7 terminator, and ApxSC structural gene (ApxSC). Recominant ApxSC was accumulated in cytoplasm of E. coli carrying pETAPX7. The expressed recombinat ApxSC was purified through column chromatographies composed of Phenyl-superose and Mono-Q , and showed the APX activity.
Genomic DNAs harboring ApxSC were isolated from a genomic library of strawberry. Restriction mapping and sequence analyses showed that a DNA is composed of a 2.5-kb of full-length ApcSC gene, a 3.7-kb of 5'-upstream region, and a 0.5-kb of 3'-downstream region. The ApxSC genomic DNA contains 10 exons and 9 introns, which is similar to the structure of pea ApxI. A primer extension analysis suggested that the transcription of ApxSC gene was started at three start sites with different degrees. The promoter region of ApxSC gene contains a sequence or structure distinct from other reported plant APX genes, though with several known functional elements such as a TATA box. Nucleotide sequences of three genomic clones were analysed and were found to show the mismatched bases. These observations were consistent with those of cDNAs that showed the mismatched three amino acid. Three genomic clones of X5, X6, and X7 corresponded to APX26, APX20, and APX11 of isolated cDNA clones. It implies that the expressions of ApxSC alleles in strawberry are codominant, due to the polyploid genome, which is supported by genomic blot and primer extension analysis.
Changes of peroxide concentration in strawberry fruit were similar with the respiration pattern in initial state (young green to mature green) and with the ApxSC expression pattern in later state (mature green to full red state). These suggest that the expression of ApxSC may be related to the perxode production during fruit ripening.
The accumulation of ApxSC mRNA during fruit ripening may protect the cells from oxidative stresses by byproducts, such as hydrogen peroxide, which are produced during ripening. Codominant and high-level expression of ApxSC in fruits may be reflected in broad ecological range of strawberry cultivar.
The regulatory mechanism of ApxSC expression stimulated in respond to during fruit-ripening remains to be tested. Hormonal treatment, the reactive oxygen species (ROS) changes, and promoter analysis to identify the functional sites could be done to accomplish the work.
딸기의 과육은 씨가 있는 Achene과 대부분의 부피를 차지하는 Receptacle로 이루어져 있으며, 발생학적으로 보았을 때, Achene이 과육이며, Receptacle은 과육이 아니다. 그러나 실제로는 과육이 아닐지라도 보통 과육이라고 불리우며, 일어나는 생리적 현상도 비슷하다고 알려져 있다. 이와 같은 딸기의 과육은 다른 조직과는 다른 특이적인 형태와 특징적인 숙성(ripening)이라는 생리적 변화를 겪는다.
이러한 딸기의 과육내에서 일어나는 특이적 현상들을 이해하기 위하여 다른 조직보다 과육에서 특이적 또는 우세하게 발현되는 단백질을 이차원 전기영동을 통하여 분리하였다. 이들을 각각 60, 40, 26, 10 KDa 단백질로 명명하였다. 이들 4 가지 단백질의 아미노 말단의 아미노산 서열을 분석해 본 결과, 26 KDa 단백질의 서열만이 분석되었다. 나머지 3종류의 단백질은 아미노 말단이 masking 되어 있는 것으로 생각된다. 26 KDa 단백질을 동정하기 위하여 결정된 아미노산 서열을 GenBank에 보고된 서열들과 상동성을 비교해 본 결과 식물 APX와 약 80%의 상동성을 가지고 있음을 알 수 있었다. 26 KDa 단백질이 APX의 일종임을 효소활성 조사에 의해 확인하였다. 따라서 이 단백질을 ApxSC로 명명하였다. 현재까지 보고된 APX유전자들의 염기서열의 비교를 통해 3종류의 oligomer를 합성하였고, 26 KDa 단백질의 결정된 아미노산 서열로부터 1종류의 oligomer를 합성하여, 이 단백질을 암호화하는 유전자분리를 위한 probe를 제작하는데 사용하였다. 합성된 4종류의 oligomer를 primer로, 딸기 과육 mRNA 를 주형으로 하여 역전사와 중합효소연쇄반응(PCR)을 수행하였다. 그 결과 크기가 다른 2종류의 산물을 얻어 염기서열을 분석해 본 결과, APX의 단편임을 확인하였다. 증폭된 PCR 산물을 probe로 사용하여 딸기 과육 cDNA library로부터 36개의 클론을 분리하였다. 한 클론의 전체 염기서열을 분석해 본 결과, 이 cDNA는 온전한 하나의 ORF를 가지고 있었으며, 상동성 비교와 분자량, transit peptide 존재 유무의 판단에 의해 식물 APX 의 여러 isoform 중 세포질(cytosol)에 위치하는 APX이었다. 분리된 cDNA의 ORF 부위와 UTR 부위를 probe로 사용하여 transcription 수준에서 조직별, 과육 발달(또는 숙성) 단계별 발현양상을 조사해 보았을 때, 분리한 ApxSC 유전자는 다른 조직보다 과육에서 우세하게 발현되는 것을 관찰할 수 있었고, 과육이 숙성됨에 따라 발현이 증가를 하여 turning 단계에서 최대치를 보이고, 그 이후로는 감소함을 보였다. 조직별, 과육 발달단계별 딸기의 활성염색을 통한APX 활성 조사는 3개의 활성을 보이는 band를 확인할 수 있었고, 그들의 발현 양상은 ApxSC의 northern blot과 비슷한 양상을 보이나, 그 양에 있어서는 차이가 있었다. 염기서열이 결정된 하나의 클론을 제외한 나머지 35개 클론의 200-300 bp의 부분 염기서열 분석은 앞의 ApxSC크론과 97-99%의 일치성을 가지는 클론이 대부분임을 확인할 수 있었다. 이러한 결과는 딸기 ApxSC가 multicopy로 존재함에 의해 또는 딸기의 다배체 특성에 의한 것으로 추정되었다. UTR 부위에서의 상동성과 Southern blot 결과는 후자일 가능성이 높음을 보여준다.
분리된 ApxSC cDNA가 기능성이 있는지를 확인하고, 항체형성을 위한 다량의 항원을 확보하기 위하여 E. coli내에서 높은 수준으로 발현시키는 체계와 순수분리하는 체계를 확립하였다.
ApxSC의 발현에 영향을 미치는 promoter 부위를 분리하기 위하여 딸기 genomic library로부터 여러 개의 클론을 분리하였다. 이 중 분석된 클론의 염기서열은 이 클론이 ApxSC의 genomic DNA와 promoter 부위를 함유하고 있음을 확인하였다. 이 DNA는 cDNA 염기서열 비교를 통해 10개의 exon과 9개의 intron을 가지고 있었다. Promoter 부위를 분석하기 위하여 먼저 primer extension analysis 통해 3군데의 transcription 개시부위를 동정하였고, 가장 보존성이 크고 개시정도가 가장 큰 부위를 +1로 명명하였다. 상위부위에 promoter에 공통적으로 존재하는 TATA box와 CAAT box가 존재하였다. 보고된 다른 promoter의 조절부위와 비교를 통해 HSE등과 같은 부위와 몇 개의 반복서열을 찾을 수 있었다. 분리된 여러 개의 클론 중 2개의 클론의 염기서열을 분석해본 결과 cDNA의 결과와 마찬가지로 분석된 부위 전체에 걸쳐 상동성이 보였다. 이러한 결과는 딸기의 다배체 특성으로 인해 ApxSC가 여러 개의 상동염색체로부터 동시에 발현된 것임을 알 수 있었다.
딸기 과육의 숙성에 따른 ApxSC의 발현변화는 숙성에 의해 증가되는 peroxides와 관련이 있음을 시사해준다. 즉, 과육의 숙성에 따라 축적된 peroxides로부터 세포를 보호하기 위한 하나의 방어 기작으로서 ApxSC의 발현이 증가된 것으로 사료된다.
