The effects of 2-acetylaminofluorene (AAF), an arylamide carcinogen, on IL-1 production and IL-1 gene expression were investigated. AAF inhibited IL-1 production in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells and rat peritonial macrophages. To further characterize the effect of AAF on the gene expression of IL-1β, we assessed the effect of AAF on the expression of IL-1β mRNA using competitive RT-PCR. Treatment of RAW 264.7 cells and mouse splenic macrophages with AAF significantly inhibited the production of IL-1β mRNA.
In order to investigate the effects of AAF on the regulation of IL-1β gene expression, the effects of AAF on the activation of transcription factors such as NF-κB, AP-1, CRE, and NF-IL6 were examined using a transient transfection and chloramphenicol acetyltransferase (CAT) assay and electrophoretic mobility shift assay. AAF inhibited the transcriptional activation of NF-κB in LPS-stimulated RAW 264.7 cells. The DNA binding activity of NF-κB was also inhibited by AAF in LPS-stimulated RAW 264.7 cells. These data suggest that inhibition of IL-1β gene expression by AAF is mediated by blocking LPS-stimulated NF-κB activation.
본 실험에서는 발암 물질로 널리 알려진 2-acetylaminofluorene (AAF)이 IL-1의 생산에 미치는 영향과 IL-1β의 유전자 발현에 미치는 영향에 대하여 알아보았다. AAF는 lipopolysaccharide (LPS)에 의해 활성화된 RAW 264.7 세포와 복강 거식세포에서 IL-1의 분비를 억제하였다. AAF가 IL-1β의 유전자 발현에 주는 영향을 알아보기 위하여 competitive RT-PCR을 이용하여 IL-1β의 mRNA 발현에 미치는 영향을 살펴보았다. RAW 264.7 세포와 생쥐 비장 거식세포에 AAF를 처리하였을 때 IL-1β의 mRNA 생성이 저해되었다.
AAF가 IL-1β의 전사 촉진의 조절에 미치는 영향을 살펴보기 위하여 전사 조절 인자인 NF-κB, AP-1, CRE와 NF-IL6의 활성에 미치는 영향을 chloramphenicol acetyltransferase assay와 electrophoretic mobility shift assay를 통하여 알아보았다. AAF는 LPS에 의해 활성화된 RAW 264.7 세포에서 NF-κB의 전사 활성을 저해하였다. LPS에 의해 활성화된 RAW 264.7 세포에서 전사 조절 인자인 NF-κB의 DNA 결합 활성 또한 AAF에 의해서 저해되었다. 이러한 사실들로부터 AAF에 의한 IL-1β의 유전자 발현 억제는 LPS에 의해 유도되는 NF-κB의 활성화를 막음으로써 일어난다는 것을 알 수 있다.
