Unnatural β-peptides adopt stable secondary structures even at short oligomers, have a wide range of structural diversity, and exhibit remarkable in vivo stability. Based on these properties, β-peptides are well documented as a building block of functional materials. In this context, it is necessary to establish a design strategy for β-peptides.
In this dissertation, helicity control of the helical β-peptide and its analogues by transforming each element consisting of the peptide backbone are discussed. Through the substitution of the oxygen atom of the amide with sulfur atoms, precise control of the secondary and higher order structure of β-peptide was achieved. Moreover, post-synthetic modification enables the synthesis of diverse derivatives. Otherwise, a helical α/γ-peptide insensitive toward chiral information was developed, and an intermediate structure in the interconversion between left- and right-handed helices was observed. In addition, enantiopure cyclic β-amino acids comprising various functional groups were successfully synthesized. These findings are expected to improve understanding of the structural properties of a diverse range of unnatural peptides and offer insights for the development of innovative peptide design strategies.
비천연 베타-펩타이드는 짧은 길이에서도 안정한 이차 구조를 이루며, 넓은 폭의 구조적 다양성을 가지고, 생체내 안정성이 우수하다. 이와 같은 특성에 기반해 베타-펩타이드를 기능성 재료의 구성 요소로 폭넓게 활용할 수 있음이 잘 알려져 있다. 이러한 맥락에서 베타-펩타이드의 디자인 전략 확립에 대한 필요성이 대두되었다.
본 학위논문에서는 나선형 베타-펩타이드와 그 유사체의 주골격을 구성하는 각각의 요소에 변화를 가해 그 나선성을 제어할 수 있음을 설명하고자 한다. 펩타이드를 구성하는 아마이드의 산소 원자를 황 원자로 치환해 펩타이드의 이차 및 고차 구조를 정밀히 조절하고, 합성 후 변환을 통해 다양한 유도체를 쉽게 합성할 수 있음을 밝혔다. 한편 카이랄 중심의 존재에도 나선형 카이랄성 선호를 보이지 않는 분자를 개발하였고, 상호 변환 과정에서의 중간 단계를 관찰할 수 있었다. 또한 폭넓은 작용기를 가져 응용 가능성이 높은 고리형 베타-아미노산을 성공적으로 합성하였다. 이 결과들은 다양한 비천연 펩타이드의 구조적 특성에 대한 이해도를 높이고, 새로운 펩타이드 디자인 전략을 수립하기 위해 활용할 수 있을 것이라 기대된다.
