The liquid-like protein condensate generated by the intracellular biomolecular phase separation phenomenon is the basic principle for the formation of various kinds of membrane-less organelles inside the cell, and the formed organelles are the basis of biological metabolic pathways and the mechanisms by which organelles play their roles. The resulting protein condensates have the advantages of free internal and external mass exchange, the ability to recruit high concentrations of biomolecules into the condensate, and specific physical properties such as liquid, gel, and solid. These characteristics of protein condensates have attracted interest in applied research through the synthesis of cell-mimicking artificial organelles outside the cell. In this paper, I designed a strategy to generate a condensate model that can modulate the dynamics of protein phase separation condensates through additional material processing and reversibly transition between liquid, gel, and solid phases. The model was able to improve the stability and duration of the condensate to external changes by modifying the internal dynamics of the condensate, and confirmed its potential as a cell-mimicking artificial organelle by confirming its biological activity inside. Next, I designed a material delivery strategy to the inside of the cell using protein phase separation condensate. Phase separation condensates were generated from different types of proteins by metal ion induction and processed directly into cells. The formation of condensates and the efficiency of material delivery to cells varied depending on the type of protein, size, charge, and time of condensate formation. The developed phase separation condensate biomolecular delivery system is simple to produce and contains high efficiency biomolecules inside the condensate. These results provide a direction for evaluating their potential as biomolecular carriers and improving their delivery efficiency.
세포 내 생체분자 상 분리 현상에 의해 생성된 액체와 같은 단백질 응축물은 세포 내부의 여러 종류의 막이 없는 세포 소기관을 형성하는 기초 원리가 되고 형성된 소기관들은 생물학적 대사 경로 및 세포 소기관들이 역할을 수행하는 메커니즘의 기초가 된다. 생성된 단백질 응축물들은 내부와 외부의 물질 교환이 자유롭고 응축물 내부로 높은 농도의 생체분자를 모집할 수 있는 장점이 있고 액체, 젤, 고체와 같은 특정 물성을 가짐에 따라 역할 수행이 구분된다. 이러한 단백질 응축물의 특징 때문에 세포 외부에서 합성을 통해 세포 모방 인공 소기관 제작을 통한 응용 연구에 관심이 집중되고 있다. 본 논문에서는 추가적인 물질 처리를 통해 단백질 상 분리 응축물의 역학을 조절하고 액체, 겔, 고체 상태로 가역적 전이가 가능한 응축물 모델 생성 전략을 설계했다. 이 모델은 응축물 내부 역학 조절로 외부 변화에 대한 응축물의 안정성과 지속시간을 향상 시킬 수 있었고, 내부에서 생물학적 활성 확인을 통해 세포 모방 인공 기관으로써의 가능성을 확인했다. 다음으로 단백질 상 분리 응축물을 사용한 세포 내부로의 물질 전달 전략을 설계 했다. 금속 이온 유도로 여러 종류의 단백질로 상 분리 응축물을 생성하고 세포에 직접 처리하였다. 또한 단백질의 종류, 크기, 전하, 응축물의 형성 시간에 따라 응축물의 형성과 세포로의 물질 전달 효율은 변화했다. 개발된 상 분리 응축물 생체분자 운반체는 생성이 간단하고 응축물 내부에 높은 효율의 생체분자를 함유한다. 이 결과를 통해 생체분자 운반체로써의 잠재력 평가와 전달 효율성을 향상 시키는 방향을 제시한다.
