This dissertation encompasses two individual research subjects. The first subject explores the correlation between folding propensity of foldamer and properties of multiscale architecture. To investigate this, three distinct α/β-residue pattern foldamers were prepared and analyzed by several characterization containing crystal structure determination, spectroscopic analyses, and computer simulation. Additionally, the assembly process of those foldamers macroscopic architectures were studied, providing insight how the folding propensity influences the physicochemical properties of those architectures. Those case studies are anticipated to contribute to broadening the perspective on understanding molecular assembly
The second chapter demonstrates structure determination of nanocrystals of metal-biomolecules using microcrystal electron diffraction (MicroED). The final refinement result exhibited good re-finement statistics comparable to those obtained by conventional X-ray diffraction method. Fur-thermore, the absolute configuration of the metal-biomolecule complex could be determined by MicoED. The successful application of MicroED for structure determination presents a promising and efficient method for solving the structure of nanocrystals composed of biomolecules.
본 학위 논문에서는 두 가지 개별적인 연구 주제를 포함한다. 첫 번째 주제는 폴더머의 접힘 성향과 멀티스케일 구조체 속성 간의 관계를 탐구힌다. 이를 위해 서로 다른 세 가지 α/β-잔기 패턴 폴더머를 합성하고 결정 구조 결정, 분광 분석 및 컴퓨터 시뮬레이션을 포함한 여러 특성 분석을 실시하였다. 또한 이러한 폴더머가 거시적 구조체로 조립되는 과정을 추적하여, 폴더머의 접힘 성향이 해당 구조체의 물리화학적 특성에 어떻게 영향을 미치는지에 대한 통찰을 제공하였다. 이러한 사례 연구는 분자 조립 이해를 넓히는 데 기여할 것으로 기대된다.
두 번째 챕터는 마이크로 결정 전자 회절 (MicroED)을 사용하여 금속-생체 분자의 나노결정 구조를 구하는 과정을 제공한다. 최종 정밀화 결과는 기존의 X선 회절 방법과 비교해봐도 견줄만한 좋은 정밀화 수치들을 나타냈다. 더욱이, MicroED를 통해 금속-생체 분자 복합체의 절대 배열을 결정할 수 있음을 밝혀냈다. 이러한 MicroED의 성공적인 응용 사례는 MicroED가 생체 분자로 이루어진 나노결정 구조를 얻기 위한 유망하고 효율적인 방법임을 제시한다.
