Discovering the cell of origin in brain tumors is challenging but important for fundamental basis of understanding the cancer-evolutionary process from beginning to terminal. Here, I identify the originating cell in human isocitrate dehydrogenase (IDH)-mutant glioma including astrocytoma and oligodendroglioma. I reveal that oligodendrocyte lineage cells containing oligodendrocyte progenitor cells (OPCs) and/or other glial cells harboring ultra-low-level of IDH1 alteration (> 0.115% of mutational burden) can be found in tumor-adjacent normal cortex in 62 patients with IDH-mutant glioma or other types of brain disease. As a biologic validation, OPC-specific somatic mutation mouse model exhibits hyperproliferative feature in mutant cells. Together, my results show that glial progenitor cell (GPC) as well as OPC that contain the IDH1 mutation are the cell of origin in IDH-mutant glioma.
뇌종양의 기원세포를 밝히는 일은 매우 어려우면서도 뇌종양 발달의 시작과 끝을 이해하기 위한 수단으로써 매우 가치 있는 일입니다. 이 연구에서 저는 인간 IDH-돌연변이 별아교세포종과 희소돌기아교세포종을 포함한 IDH-돌연변이 신경교종의 기원세포를 밝혔습니다. 저는 62명의 뇌종양 환자의 종양 주변 뇌피질 조직에서 희소돌기아교세포 전구세포를 포함한 희소돌기아교세포 계통 세포와 별아교세포를 포함한 교세포들 가운데 초저빈도 (0.115% 수준) 로 존재하는 IDH1 체성 돌연변이를 발견했습니다. 생물학적 검증의 일환으로 저는 희소돌기아교세포 전구세포에서만 특이적으로 종양유발돌연변이를 만들어서 이 돌연변이 세포들이 과증식하는 현상을 마우스에서 관찰하였습니다. 즉, 제가 이 연구에서 확인한 결과에 따르면, IDH1 돌연변이를 획득한 희소돌기아교세포 전구세포를 포함한 교세포 전구세포가 IDH-돌연변이 신경교종의 기원세포입니다.
