MicroRNA has been recognized as an important biomarker for various diseases, including cancer, and has been applied in early-stage diagnosis, therapeutic, and biomedicine. However, current methods for microRNA profiling and imaging are limited for temporal analysis, due to essential processes such as microRNA extraction or cell fixation. Therefore, a molecular probe capable of detection in living cells is necessary. In this thesis, I proposed an imaging platform designed for quantification of dynamic intracellular levels of microRNA. I investigated effectiveness of strategies, encompassing microRNA assays, transfection carriers, and ratiometric analysis techniques. The single-labeled probe was employed to minimize cytotoxicity arising from organic dyes and to reduce batch-to-batch variations in labeling. This approach is based on photoinduced electron transfer between guanine and FAM. The innovative design, where five guanines are placed adjacent to the fluorescent dye regardless of the target, ensures efficient and consistent quenching. The targeted microRNA specifically binds to hairpin DNA, and the subsequent cascade of toehold-mediated strand displacement releases the target, allowing for recycling and signal amplification. A single base-pair mismatch in the stem of the hairpin DNA boots the target-triggered strand displacement, resulting in an over 20-fold increase in sensitivity and achieving a pM level of limit of detection. The designed hairpin DNAs, encapsulated within the interlayers of multilamellar liposome vesicles, was delivered into cells with a high transfection yield, maintaining high stability. The microRNA imaging platform successfully monitored changes in microRNA levels in living cells over several hours of drug treatment. This long-term microRNA imaging system have great potential in biomedical field.
마이크로 RNA는 암을 포함한 다양한 질병에서 중요성을 인정받은 바이오마커로, 초기 진단 및 치료, 약학 등의 다양한 분야에서 활용되어 왔다. 하지만 기존 마이크로 RNA 발현 분석 및 이미징 기법은 마이크로 RNA 추출 및 세포 고정과 같은 과정이 필수적이므로 시간에 따른 분석에는 한계가 있다. 따라서 생세포 내에서 구동이 가능한 분자 프로브가 필수적이다. 본 학위 논문에서는 세포 내 마이크로 RNA의 양 변화를 지속적으로 감지할 수 있는 이미징 플랫폼을 제안했다. 효과적인 이미징을 위해 마이크로 RNA 어세이, 전달 구조체, 비율 계량 측정법을 아우르는 여러 전략의 효용성을 다루었다. 유기염료로 인한 세포 독성 및 배치 간 표지 편차 최소화를 위해 단일 유기염료 표지 방식의 프로브가 채택되었다. 이는 구아닌과 FAM 간의 광유도 전자 전달을 기반으로 하며, 타겟과 상관없이 다섯개의 구아닌이 발광 유기염료 주변에 위치하는 새로운 디자인을 통해 효율적이고 일관된 광유도 전자 전달이 일어날 수 있도록 한다. 타겟은 헤어핀 DNA와 특이적으로 결합하고 이후 연속적인 토홀드 매개 가닥 변위 반응에 따라 최종 산물에서 방출되어 재활용되면서 시그널을 증폭한다. 헤어핀 DNA 줄기의 염기쌍 불일치는 타겟 마이크로 RNA에 의한 가닥 변위 반응을 촉진하며, 이에 따라 민감도가 20배 이상 증가하여 피코몰 농도 수준의 검출한계를 보인다. 개발된 헤어핀 DNA 프로브는 다층 라멜라 지질 소포체의 층사이에 포집되어 매우 높은 효율로 높은 안정성을 유지하며 셀 내부로 전달된다. 마이크로 RNA 이미징 플랫폼을 이용하여 수 시간 동안의 항암제 처리에 따른 살아 있는 암세포 내의 마이크로 RNA의 변화를 성공적으로 확인하였다. 제안된 마이크로 RNA 이미징 시스템은 생체 의학 분야에서 큰 잠재력을 가지고 있다.
