The cell membrane is a thin lipid bilayer composed of various phospholipids and proteins, consisting of laterally segregated domains with different chemical compositions and physical properties. The dynamic organization of membranes is crucial for a variety of biological functions and is closely related to the interaction of cell membranes with various biomolecules, including proteins, peptides, and drug molecules. This thesis aims to analyze the reorganization of lipid membranes induced by proteins and peptides using freestanding planar lipid membranes. Through real-time visualization of membranes, we tried to understand how interactions with proteins or peptides can rearrange membrane lipids, and how this could be related to their biological functions. First, we studied protein condensation that occurs on the surface of lipid membranes. By enabling protein binding on both sides of the lipid membrane, we observed a new phenomenon, the transmembrane coupling of protein condensates, which is mediated by the lateral rearrangement of the lipid membrane. Second, using phase-separated membranes that mimic the lateral heterogeneity of cell membranes, we examined the dynamic reorganization of the membranes by the arginine-rich cell-penetrating peptide. Furthermore, by analyzing the membrane penetration of the peptide, we found that cell-penetrating peptides can actively facilitate their membrane translocation by fluidizing the membranes. Our findings reveal a new mechanism for how protein condensates or cell-penetrating peptides act on cell membranes, and we expect that our work will also offer valuable insights for exploring the dynamic membrane remodeling induced by membrane-biomolecule interactions, which could be responsible for a variety of biological processes.
세포막은 다양한 인지질과 단백질로 구성된 얇은 인지질 이중층으로, 조성 및 성질이 다른 작은 구분된 영역들이 불균일하게 분포하고 있다. 지질막의 능동적인 측면 구조 변화는 다양한 생물학적 기능에 필수적인 역할을 하며, 이는 단백질, 펩타이드, 약물분자 등 다양한 생체분자와 세포막 간의 상호작용과도 밀접한 연관이 있다. 본 학위 논문에서는 자립형 평면 지질 이중막 시스템을 사용하여 단백질 및 펩타이드에 의해 유도된 지질막의 재구성을 분석하는 것을 목표로 한다. 막의 실시간 가시화를 통해 우리는 단백질이나 펩타이드와의 상호작용이 어떻게 막 지질을 재배열할 수 있는지, 그리고 이것이 생물학적 기능과 어떻게 관련될 수 있는지 이해하고자 했다. 첫번째로 액체-액체 상분리가 가능한 단백질을 사용하여 막 표면에서 일어나는 단백질 응축을 연구했다. 우리는 지질막 양쪽 표면에 모두 단백질의 결합이 가능하게 하여 이전에 목격된 적 없는 지질막 재배열을 매개로 한 단백질 응축물의 막 횡단 커플링 현상을 관찰할 수 있었다. 두번째로, 세포막의 이질성을 모사하는 상 분리된 지질막을 사용하여 아르기닌이 풍부한 세포 투과 펩타이드에 의한 지질막의 동적 재구성을 조사했다. 또한, 펩타이드의 막 투과 분석을 통해 세포 투과 펩타이드가 막을 유동화 함으로써 적극적으로 막 투과를 촉진할 수 있음을 발견했다. 우리의 연구 결과는 단백질 응축물 및 세포투과펩타이드가 세포막에서 어떻게 작용하는지에 대한 새로운 메커니즘을 밝혀 냈을 뿐 아니라, 다양한 생물학적 과정에서 발생하는 막-생체분자 상호작용에 의한 동적 지질 재배열을 탐구하는 데 중요한 통찰력을 제공할 것으로 기대한다.
