Tau protein found in neurofibrillary tangle inside of a neuron, which is one of two conventional hallmarks of Alzheimer’s disease has been receiving large attention recently. Normal Tau is primarily known to bind to microtubules, an anionic cytoskeleton by electrostatic interaction. It is difficult to observe the conformation of Tau because it has almost no secondary or tertiary structure, so-called an intrinsically disordered protein (IDP) like a random coil. In addition, it is hard to predict the conformation of Tau, a polyampholyte that has both positive and negative charges. Therefore, the molecular structure and mechanism of Tau binding to microtubules are not yet elucidated. The main goal is to build a model of the association between Tau and microtubules and analyze the mechanism by monitoring structural changes of microtubules in the presence of Tau under ionic regulation. The hypothesis is that electrostatic interaction leads to the structural changes of Tau and microtubules. The Tau-microtubule bundles under controlled ionic strength and osmotic pressure were monitored and analyzed by small-angle X-ray scattering (SAXS), transmission electron microscopy (TEM), confocal laser scanning microscopy (CLSM), and western blot immunoassay techniques. Here, we suggest that the intermolecular structures of Tau bound to microtubules and changes in the physical property of Tau proteins under ionic regulation.
타우 단백질은 알츠하이머병의 핵심 물질로 최근 주목 받고 있다. 알츠하이머병의 전통적인 두 가지 특징 중 하나로 신경 세포 내부에서는 비정상적인 타우가 모여 신경섬유 형태로 얽힌 구조가 관찰된다. 정상적인 타우는 세포 골격을 이루는 마이크로튜불에 정전기적 상호작용에 의해 결합하여 구조를 안정화한다고 알려져 있다. 타우는 양전하와 음전하를 모두 가지고 있으며, 정해진 구조가 없는 미접힘 단백질이기 때문에 구조를 관찰하고 분석하기 어렵다. 이로 인해, 타우와 마이크로튜불의 결합 구조와 메커니즘은 아직 명확히 밝혀져 있지 않다. 본 연구에서는 타우와 마이크로튜불의 정전기적 상호작용에 영향을 미치는 이온 세기를 조절하여 마이크로튜불의 구조 변화를 관찰하고, 이로부터 타우의 결합 구조 모델을 구축하고, 메커니즘을 분석하고자 했다. 소각 X선 산란, 투과 전자 현미경, 공초점 레이저 주사 현미경, 웨스턴 블롯 기법으로 마이크로튜불 다발에서 타우의 역할과 구조, 그리고 이온 세기에 따른 타우의 물리적 특성 변화를 제시하였다.
