Lipid vesicles are self-assembled structures composed of lipid bilayers that surround water or cellular cytoplasm. The electric double layers around lipid vesicles play crucial roles in various fields, such as artificial cells and drug delivery systems. While previous studies have reported the controllable adsorption of ions onto the membrane surfaces depending on the lipid composition, systematic researches on the overall structure of the electric double layer, including ion distributions and effective surface charges, have been lacking. In this dissertation, we measure the second virial coefficient (B$_2$) of lipid vesicles using optical bottle technique and subsequently profile the characteristics of the electric double layer, including charge reconstruction factors and Debye length. In particular, we demonstrate that the involvement of water-derived H$^+$ ions is a significant factor influencing charge regulation, and the presence of DOPC around DOPG can induce giant charge reconstruction of the lipid vesicles by altering the topological structure of the stabilization shell for adsorbed cations.
지질 소포는 지질 이중층이 물이나 세포질을 둘러싸고 있는 자가조립구조이다. 지질 소포 주변의 전기 이중층은 인공세포나 약물 전달 시스템과 같은 다양한 분야에서 중요한 역할을 한다. 기존 연구들에서는 지질 종류에 따라 막 표면에 이온이 흡착되는 것을 조절할 수 있다고 보고되었지만, 이온 분포와 유효 표면전하와 같은 전반적인 전기 이중층 구조에 대한 체계적인 연구들은 여전히 부족한 상태였다. 본 학위 논문에서는 광학 병 기술을 통해 지질 소포의 2차 비리얼 계수 (B$_2$)를 측정하고, 그로부터 전하 재구축 인자 및 디바이 길이와 같은 전기 이중층의 특성을 프로파일링한다. 특히, 물에서 유래된 H$^+$ 이온의 개입이 전하 조절에 있어서 주요한 영향을 미치는 요인 중 하나라는 사실과 더불어, DOPG 주변의 DOPC의 존재가 흡착된 양이온 주위 안정화 껍질의 위상학적 구조를 바꿈으로써 지질 소포의 대규모 전하 재구축을 일으킬 수 있음을 보이고자 한다.
