Carboxylic acids are crucial metabolites for understanding metabolic disorders and disease diagnosis. In this work, we developed selective $^{19}F$ labeling protocol for carboxylic acids in complex biofluids, employing pivaloyl chloride as an amide coupling reagent. By masking reactive functional groups such as thiols, amines, and alcohols, it enables the specific fluorine tagging of carboxylic acids, those selectively activated to the acid anhydrides. We examined the $^{19}F$ chemical shifts of 79 carboxylic acids and constructed the fingerprint region. Based on this newly developed method, we achieved simultaneous identification of carboxylic acid containing metabolites in human plasma and successfully quantified both small organic acids and fatty acids. This novel analytical method shows great potential for gaining valuable insights into the metabolic processes of living organisms.
카복실산은 대사 장애와 질병 진단에 연관되어 있는 중요한 대사 물질이다. 본 학위논문은 복잡한 생체 유체에 존재하는 카복실산에 선택적으로 불소를 표지하는 방법에 대해 기술하였다. 싸이올, 아민, 알코올과 같은 반응성이 큰 작용기를 비활성화시키고 카복실산은 산 무수물로 선택적으로 활성화시킴으로써 카복실산의 선택적인 $^{19}F$ 표지를 가능하게 하였다. 79개 카복실산의 $^{19}F$ 화학적 이동을 검토하여 지문 영역을 구축하였으며, 이 분석 방법에 기반하여 사람의 혈장에 존재하는 카복실산의 동시 분석을 수행하고 유기산과 지방산의 농도를 성공적으로 정량하였다. 이러한 분석 방법은 유기체의 대사 과정을 이해하는 데 활용될 수 있다.
