Engineering of bacterial strains with efficient antibody secretion is important in terms of economical benefits compared to that of eukaryotic cells. However, the work of strain design for fine antibody secretion has been constrained for further development by several reasons such as structural stability of antibody and safety issue of host cell. Especially, even though Corynebacaterium glutamicum has remarkable characteristics that it is a GRAS (Generally Regarded as Safe) strain that has its thick membrane system to maintain intracellular condition, there is a lack of study on the strain with antibody secretion. Thus, for further development of study in antibody secretion from bacterial strain, this study was based on C. glutamicum, and in purpose of excluding the problem on structural stability of antibody, nanobody antibody called shark VNAR antibody was used to investigate the potential of C. glutmaicum as a host cell for antibody secretion. For contributing a condition of selecting diverse signal peptides to this study, an artificial signal peptide library was constructed upstream of gene encoding a humanized shark VNAR antibody called HuVAR version I. FAST (Flourescene-activating and Absorption-Shifting Tag) dye assay was employed for efficient screening of noble strains with secretion capabilities. The sorted strains were further confirmed their secretion from 50ml BHI medium cultivation. Overall, five remarkable strains has been discovered with different signal peptides and secretion abilities was confirmed in terms of protein expression.
진학 생물보다 더 경제적인 이득을 취할 수 있기 때문에 효율적인 항체 분비 능력을 박테리아 균주 개량은 중요하게 여기고 있다. 그러나 균주 안전성 문제와 세포에서 분비된 항체의 구조적 안정성 문제와 같은 여러 이유로 인해 균주 개량 작업은 더 나은 발전을 이루지 못 하고 있다. 특히 코리네박테리움 글루타미쿰 균주는 그라스 균주이며 두꺼운 외막 시스템으로 인해 세포 내 환경을 변함없이 유지할 수 있다는 특징을 가졌지만, 항체 분비에 대한 연구는 많이 이루어지 않았다. 본 연구에서는 박테리아 균주의 항체 분비 연구가 발전될 수 있도록 코리네박테리움 글루타미쿰을 기반으로 항체 분비 연구를 진행했었다. 항체의 구조적 안정성 문제를 최대한 배제하기 위해 상어 유래 단일 도메인 항체 기반의 나노바디 항체를 이용했으며 코리네박테리움 글루타미쿰이 항체 분비 호스트 생물으로써의 잠재력을 가지는지 조사하였다. 본 연구에서는 다양한 신호 펩타이드들을 선별할 수 있는 환경을 조성하기 위해 인간화된 상어 나노바디 항체 버전 I에 신호 펩타이드 라이브러리를 인위적으로 구축하였고, 뚜렷한 분비 능력을 나타내는 균주들을 선별할 수 있도록 흡수 파장으로 인해 형광도 활성화되는 태그 단백질(FAST)을 기반으로 한 염료 분석을 이용하였다. 그리고 선별된 균주들은 50ml BHI 배지 배양에서 추가로 확인해서 확실하게 나노바디 항체가 분비되는 것을 검증하였다. 결과적으로 본 연구에서 5개의 새로운 인공 신호 펨타이드가 발견되었고 단백질 발현까지 확인하였다.
