The immune cells play a crucial role in maintaining homeostasis within tissues, constantly monitoring the surrounding environment and responding to external changes. Microglia, the immune cells in the brain, undergo changes in shape and increased motility when activated by external stimuli. This suggests the significance of the motility of microglia in the brain's immune response. Recent research has revealed that the stiffness of the brain varies depending on the region, aging process, and the progression of neurodegenerative diseases such as Alzheimer's. Therefore, this study aims to gain deeper insights into the brain's immune response by investigating how changes in brain stiffness affect the motility of microglia. Firstly, we propose a method using both phase-contrast microscopy and differential interference contrast microscopy to simultaneously decompose the motility and shape changes of microglia. This allows for accurate measurement of both the motility and shape changes of microglia. Through this, we confirmed the effect of the brain substrate stiffness on the motility of microglia. The results revealed differences in the diffusivity, directional switching tendency, and traction force of microglia depending on changes in brain substrate stiffness. These findings indicate that the stiffness of the brain plays a crucial role in the motility of microglia, and the resulting changes in the physical motility characteristics of microglia have the potential for use as a biomarker for the brain's immune response.
면역 세포는 조직 내 항상성을 유지하는데 중요한 역할을 하며, 주변 환경을 지속적으로 감시하고 외부 변화에 반응한다. 뇌의 면역 세포인 미세아교세포는 외부 자극에 의해 활성화되면 모양이 변화함과 동시에 운동성이 증가한다. 이는 뇌 내 면역 반응에 미세아교세포의 운동성이 중요한 역할을 한다는 것을 시사한다. 최근 연구에서 뇌의 강성이 뇌의 부위, 노화 과정, 그리고 알츠하이머 병과 같은 신경퇴행성 질환의 병기에 따라 변화한다는 것이 밝혀지고 있다. 따라서 본 연구에서는 뇌의 강성 변화가 미세아교세포의 운동성에 어떤 영향을 미치는지를 연구함으로써 뇌의 면역 반응에 대한 더 깊은 통찰력을 얻는 것을 목표로 한다. 먼저 위상차 현미경과 미분간섭 현미경을 동시에 사용해 미세아교세포의 운동성과 형태 변화를 분해하여, 미세아교세포의 운동성과 세포의 형태 변화를 동시에 정확하게 측정하는 방법을 제시한다. 이를 통해 뇌의 기질 강성에 따른 미세아교세포의 운동성을 확인하였으며, 그 결과 뇌의 기질 강성이 달라짐에 따라 미세아교세포의 확산성, 방향 전환 경향성, 견인력에 차이가 있음을 밝혔다. 이러한 결과는 뇌의 강성이 미세아교세포의 운동성에 중요한 역할을 하며, 이에 따른 미세아교세포의 물리적인 운동 특성 변화가 뇌 내 면역 반응의 바이오마커로써의 잠재적인 사용 가능성이 있음을 시사한다.
