Sodium is one of the most important micronutrients for the maintenance and survival of living organisms. Animals primarily detect sodium through the action of peripheral taste cells, which evokes an unique salt taste. However, it is poorly unknown whether animal respond to sodium in the digestive tract that independently of taste sensing. Here we report drosophila that unable to detect or tract sodium became capable of responding to sodium after a period of dietary sodium deprivation. From a screen for cells required for the deprivation-induced sodium preference, we identify a population of anterior enteric neurons, which we named INSO (Internal Sodium Sensing) neurons, that are necessary and sufficient for inducing behavioral preference for sodium following deprivation and marked with AstC-R1. INSO enteric neurons innervate into the gut epithelia mainly through their dendritic processes and send their axonal projection to the brain and crop duct. INSO enteric neurons respond immediately and specifically to sodium ion. We found the ligand AstC has putative role that mediate NaCl preference by modulating AstC-R1+ enteric neurons and AstC was expressed in posterior midgut enterocytes response to sodium ingestion. Together, this study identifies a previously unknown taste-independent sodium sensor that is essential for the maintenance of sodium homeostasis.
나트륨은 가장 중요한 부영양소 중 하나로서 생물의 생존 및 균형 유지에 관여한다. 동물들은 대체로 짠맛을 통해 맛 감지 세포 등의 말초 감지 세포에서 나트륨을 감지한다. 하지만, 이 맛에 대한 감지와 독립적으로 나트륨에 대한 감지 기작은 많이 알려져 있지 않다. 본 학위 논문에서는 초파리에서 맛 감지기관과 별개로 나트륨 박탈에 따라 나트륨을 찾아가는 현상을 발견하였다. 이에 어떠한 세포가 이에 관여하는지 찾아보았고, 그 결과, 장 앞 부분에 존재하는 신경세포 (INSO) 가 이를 조절하는 것을 발견하였고 AstC-R1 이라는 유전자를 발현하는 것을 확인하였다. 이 신경세포들은 장에 수상돌기를, 뇌와 식도에 축삭돌기를 뻗고 있었고, 즉각적으로 나트륨을 감지 한다는 것을 확인하였다. 또한 이의 리간드인 AstC는 나트륨에 반응하여 장 후반부에서 발현하여 장신경세포를 조절해 소금에 대한 선호를 조절하는 잠재적 역할을 할 수 있다는 것을 발견하였다. 이를 통해 우리는 지금까지 밝혀지지 않은 초파리의 장 내에서 맛 감지 기작과 별개의 또 다른 나트륨에 대한 감지기작을 밝혀내었다.
