Ultrasound brain stimulation is a promising next-generation technology for the treatment of brain disorders because of its high stimulation resolution compared to other non-invasive brain stimulation methods. Evaluating the efficacy of ultrasound brain neuromodulation is a critical, yet challenging step required for clinical trials and commercialization. However, conventional ultrasound devices used for preclinical systems are currently too bulky to target brains of mice, the most commonly used animal model for brain disorders. In this thesis, I developed a miniaturized ultrasound neuromodulation system composed of a capacitive micromachined ultrasound transducer (CMUT) and an acoustic lens designed based on the phase-conjugation mirror principle which compensates for the skull aberration to achieve accurate brain targeting. The advantages of using CMUT include small size of the system and uniform acoustic phase from the transducer. Moreover, focused ultrasound stimulation in a freely-moving mouse was demonstrated for the first time using the developed system. By enabling the evaluation of efficacy and safety of ultrasound neuromodulation in small animals, the developed system can be used in a wide range of applications such as identification of treatable brain disorders, understanding the mechanism of neuromodulation, and investigating the long-term efficacy and side-effects of ultrasound neuromodulation.
초음파 뇌자극 기술은 다른 뇌자극 기술에 비교하여 높은 공간해상도의 자극이 가능하여 뇌질환 치료의 유망한 차세대 기술이다. 초음파 뇌자극의 효능을 검증하는 것은 임상적인 시도나 상용화를 위해서 꼭 필요하면서도 어려운 단계이다. 그러나 기존에 임상적으로 사용되던 경두개 초음파 집속 장치는 뇌질환 동물모델로 널리 사용하는 생쥐에 그대로 적용하기에는 너무 거대하였다. 본 학위논문에서는 정전용량식 초음파 탐촉자(CMUT) 및 두개골의 영향을 보정하여 정확한 뇌 타겟팅을 위해 음향결합반사경 원리로 설계한 음향렌즈를 결합하여 초소형 경두개 초음파 집속 장치를 개발하였다. CMUT을 사용하여 얻을 수 있는 장점은 작은 시스템 크기와 균일한 음향 위상이었다. 또한 개발한 시스템을 이용하여 자유롭게 움직이는 생쥐에서 집속 초음파 뇌자극을 최초로 보였다. 소동물에서 초음파 뇌자극의 효능과 안전성의 검증을 가능케 하는 본 시스템은, 치료 가능한 뇌질환 탐색, 뇌자극의 메커니즘 이해, 초음파 뇌자극의 장기 효과 및 부작용 연구 등 넓은 범위에 응용될 수 있다.
