Focused ultrasound stimulation (FUS) is a promising therapeutic modality for noninvasive targeted neuromodulation. Consequently, there is a growing need to investigate the direct neuronal effects of FUS using various imaging techniques. Functional magnetic resonance imaging (fMRI) is a well-established and mature technology that is theoretically compatible with FUS and is capable of whole-brain monitoring in real-time. However, existing ultrasonic stimulation systems are incompatible with high-resolution fMRI (> 7T) for small animals because commercially-available ultrasound transducers are generally bulky and cause external image artifacts under high magnetic field strength. This paper reports a miniaturized, MR-compatible ultrasound transducer system for simultaneous monitoring of FUS-induced acoustic effects using high resolution (9.4 T) fMRI in mice. Our system integrated (1) MR-compatible materials, (2) robust packaging and interface designs for ultrasound transmission, and (3) electromagnetic noise reduction techniques. Using this system, we demonstrate artifact-free echo-planar imaging (EPI) signal changes due to ultrasound brain stimulation in vivo. Our results provide a critical demonstration of miniaturized MR-compatible ultrasound stimulation systems for in vivo studies in small animals usinghigh-resolution functional imaging.
초음파 자극은 비침습적으로 표적 신경을 조절할 수 있는 유망한 치료 양식이다. 따라서 다양한 영상 기술을 사용하여 초음파 자극의 직접적인 신경 효과를 조사할 필요성이 증가하고 있다. 특히, 기능적 자기공명영상(fMRI)은 이론적으로 초음파 자극과 호환 가능하며 실시간으로 전체 뇌를 모니터링할 수 있는 확립된 기술이다. 그러나 기존의 상용 초음파 트랜스듀서는 부피가 크며, 높은 세기의 자기장 내에서 영상 결함을 유발하기 때문에 소동물용 고해상도 fMRI(< 7T)와 호환 가능하지 않았다. 본 학위 논문에서는 초소형의 자기공명영상 장치와 호환 가능한 초음파 자극 시스템을 제작하여, 높은 해상도의 생쥐용 fMRI (9.4 T)에서 초음파에 의한 효과를 실시간으로 모니터링할 수 있었다. 이 시스템에서는 (1) 자기공명영상장치와 호환 가능한 재료, (2) 초음파 전달을 위한 견고한 패키징 및 경계면, (3) 전자기 잡음 감소 기술을 통합하였다. 생체 내 초음파 뇌 자극으로 인한 에코 평면 이미지의 결함이 없음을 이 시스템을 사용하여 입증하였다. 이 연구 결과는 자기공명영상장치와 호환 가능한 초소형 초음파 자극 시스템을 통해 고해상도 기능 이미징을 기반으로 한 소동물 생체 내 연구의 가능성을 보여준다.
