After the declaration of a global pandemic caused by the COVID-19, the development of therapeutic drugs using inhalation delivery of mRNA is drawing a huge attention. Currently, the most widely known mRNA carriers are ionizable lipid nanoparticles (LNPs), which compensate for the shortcomings of previously known cationic lipid particles. However, because of the structural instability coming from shearing stress of aerosolization process and the disturbance of the pulmonary surfactant layer in the human body, papers have been continuously published that LNP is not efficient in delivering mRNA into the lungs. Therefore, in this study, we develop an mRNA-liposome complex based on animal-derived pulmonary surfactant that can deliver mRNA into the lungs and overcome the disadvantages of the existing LNPs.
코로나 바이러스에 의한 전 세계적 팬데믹 선언 이후 mRNA의 흡입 전달을 활용한 치료제 개발이 주목을 받고 있다. 현재 가장 널리 알려진 mRNA 전달체는 이온화성 지질 나노입자(LNP)로, 기존에 알려진 양이온성 지질 입자들의 단점을 보완했다. 하지만 네뷸라이저에서 에어로졸화 되는 과정에서 오는 전단력에 의한 구조적 불안정성, 그리고 인체 내의 폐 계면활성제층에 의해 기존의 LNP는 mRNA의 폐 내 전달에 효율적이지 못하다는 연구 결과가 지속적으로 발표되었다. 따라서 이 연구에서는 동물 유래의 폐 계면활성제를 기반으로 한 mRNA-리포좀 복합체 입자를 개발함으로써 기존의 LNP가 가지는 단점을 보완하고 mRNA를 폐 내로 전달할 수 있는 보편적인 전달 시스템을 구축하고자 한다.
