Severe acute respiratory syndrome-coronavirus 2 (SARS-CoV-2), targeting human alveoli, caused a coronavirus disease 2019 pandemic (COVID-19), and multiple viral variants of the virus, such as Alpha, Delta, and Omicron, continuously emerged. However, the lack of models mimicking the cellular physiology of hAT2 cells and systems to simultaneously compare the infectivity of multiple viral variants limits understanding of infection dynamics. I aim to build systems of (1) understanding pathogenesis of the virus and (2) comparing the infectivity of multiple viral variants. In the first part, I established a human lung alveolar organoid infection model for SARS-CoV-2. With successful establishment of alveolar organoids, I infected the organoids with the virus and demonstrated robust infection through imaging-based and transcriptome- analysis. I revealed rapid viral replication during 24 hours after infection and robust innate immune response, including interferon-associated and proinflammatory genes in alveolar organoids. In addition, using unique mutation, I identified a single viral entry can fully infect an alveolar cell. I established human physiology mimicking infection model and it could be applied to investigate the pathogenesis of SARS-CoV-2. In the second part, I integrated human alveolar organoids and single-cell transcriptome sequencing to compare relative infectivity of multiple viral variants. As a proof-of-concept study, I chose four highly transmissible SARS-CoV-2 variants, including GR (B.1.1.119), Alpha (B.1.1.7), Delta (B.1.617.2), and Omicron (BA.1). With unique genomic mutations of each viral variant, a rapid evaluation of the relative infectivity in controlled experiment was possible. My system demonstrated that the Omicron variant is 5- to 7-fold more infectious to human alveolar cells than the other SARS-CoV-2 variants at the initial stage of infection. To my knowledge, the system measures the relative infectivity under multiple virus competition and provides new experimental procedures monitoring newly emerging viral variants.
제 2형 중증급성호흡기증후군 코로나바이러스는 사람 허파꽈리를 감염시켜 코로나바이러스감염증 -19을 일으켰고, 알파, 델타, 그리고 오미크론과 같은 변이 바이러스는 계속 출현했다. 하지만, 사람 폐포 제2형 세포 생리를 잘 반영하는 실험 모델과 여러 변이 바이러스의 감염력을 동시에 비교하는 시스템의 부재로 감염의 역학을 이해하는데 한계가 있었다. 이 연구에서 (1) 바이러스 의 병리기전을 이해하고 (2) 여러 변이 바이러스의 감염력을 비교하는 모델을 만들고자 했다. 첫 부분에서, 본 연구에서 사람 폐포 오가노이드를 이용하여 제 2형 중증급성호흡기증후군 코로나바 이러스 감염모델을 만들었다. 성공적인 폐포 오가노이드의 설립 후, 오가노이드에 바이러스를 감 염시키고 이미징과 전사체를 이용하여 강력한 감염을 확인하였다. 처음 24시간 동안 바이러스의 빠른 복제와 3일 차의 강한 선천 면역 반응을 관찰하였다. 그리고 바이러스의 고유 돌연변이를 이용하여 하나의 바이러스가 하나의 세포를 완전히 감염시킬 수 있음을 입증하였다. 본 연구는 사람 생리를 반영하는 감염모델을 만들었고 제 2형 중증급성호흡기증후군 바이러스의 병리기전을 이해하는데 도움을 줄 것으로 예상한다. 두 번째 부분에서, 사람 폐포 오가노이드와 단세포 전장 전사체 시퀀싱을 이용하여 여러 바이러스의 상대적 감염도를 측정하고자 했다. 개념 증명을 위 해, 우리는 감염력이 높은 변이 바이러스 4개 (GR (B.1.1.119), 알파 (B.1.1.7), 델타 (B.1.617.2), 그리고 오미크론 (BA.1))를 골랐다. 각 변이 바이러스의 고유 돌연변이를 이용해서 통제된 실험이 가능함을 보였고, 오미크론이 동일 감염력의 경우에 비해 5-7배 강한 감염력을 보 였다. 이 방법은 새로 등장하는 바이러스 변이의 감염력을 측정하여 새로운 우려변이를 감시할 수 있을 것으로 예상한다.
