Colorectal cancer and gastric cancer are cancers with high incidence and mortality rates worldwide. Although surgical and chemical therapies are commonly used, there is still a need for research on effective therapeutic targets. Recent studies demonstrated that EHMT2, a histone methyltransferase, plays a role in the proliferation and metastasis of various cancers, including breast cancer, liver cancer, and lung cancer. In this study, we confirmed that propionate, a metabolite of gut microbiota, induces apoptosis of colorectal cancer cells. We revealed that the induction of apoptosis by propionate is mediated by the degradation of EHMT2, which is highly expressed in colon cancer, through poly-ubiquitination induced by the upregulation of HECTD2, an E3 ubiquitin ligase. We also showed that the inhibition of EHMT2 directly increases the expression of TNFAIP1, leading to apoptosis of colorectal cancer cells. In addition, we found that inhibiting EHMT2, which is highly expressed in gastric cancer, induces G1 cell cycle arrest and inhibits the growth of gastric cancer cells by increasing the expression of TP53INP1, a direct target gene of EHMT2. We confirmed this effect using 3D culture system and patient-derived gastric cancer organoids. Therefore, we suggest that EHMT2 can be an effective target for the treatment of colorectal and gastric cancers.
대장암과 위암은 세계적으로 높은 발병률과 사망률을 보이는 암이다. 외과적 수술과 화학적 약물에 의한 치료법이 보편적으로 사용되고 있지만 여전히 효과적인 치료 표적의 연구가 필요한 상황이다. EHMT2는 히스톤 메틸화효소로서 유방암, 간암, 폐암과 같은 다양한 암의 생장과 전이에 영향을 준다는 연구가 이루어져있다. 우리는 이번 연구에서 장내 미생물의 대사체인 프로피오네이트가 대장암 세포의 세포사멸을 유도한다는 것을 확인했다. 프로피오네이트에 의해 유도되는 세포사멸이 대장암에서 높은 발현을 보이는 EHMT2의 단백질분해를 통해 유도됨을 밝혔고 이러한 단백질분해는 E3 유비퀴틴 연결효소인 HECTD2가 프로피오네이트에 의해 증가함으로서 일어남을 밝혔다. 또한 EHMT2의 단백질분해를 통해 TNFAIP1의 발현을 증가시켜 대장암세포의 세포사멸을 유도함을 밝혔다. 다음으로 위암에서 높은 발현을 보이는 EHMT2를 저해했을 때 표적 유전자인 TP53INP1의 증가를 통해 세포주기 G1의 정지가 유도되어 위암세포주의 생장을 억제함을 밝히고 이러한 효과를 3차원 세포배양기술과 위암 환자 유래 오가노이드를 이용하여 확인하였다. 따라서 우리는 EHMT2가 대장암과 위암의 치료에 효과적인 표적이 될 수 있음을 제시한다.
