The study of the relationship between structure and function in neuronal networks is an important area of neuroscience. Neuronal networks are known to have structural features at different shapes and scales, which have a significant impact on their function. In particular, modularity has received much attention as a structural feature that affects the robustness to stimulation, survivability, and functional diversity of neuronal networks. In addition, alginate hydrogel can be erased in the middle of the culture period with minimal damage to the neuronal networks, allowing the structure of the networks to be manipulated. In this study, I aimed to create neuronal network models with clustered structures using alginate to reproduce the modularity of neural networks, and I modulated their connections and developed a functional analysis system. For the functional analysis of neuronal networks, I first developed a cross-correlation-based connectivity analysis method and improved network burst motif finding method. I also implemented an automatic analysis system to apply them. The clustered networks were then cultured on microelectrode array using alginate hydrogel patterning, and their functional properties were analyzed. They showed differences in connectivity and motif properties compared to unstructured neuronal networks. The combination of chemical patterning and clustered networks was used to modulate network connectivity. The clustered networks created through linear and modular patterns showed not only structural connections but also functional connections based on the patterns.
신경 네트워크의 구조와 기능 사이의 연구는 뇌과학에서 중요한 분야이다. 신경 네트워크는 다양한 형태와 스케일에서 구조적 특징을 가지는 것으로 알려져 있는데, 이들은 기능에 상당한 영향을 끼친다. 특히 모듈라리티는 신경 네트워크의 자극에 대한 안정성, 생존율, 기능적 다양성 등에 영향을 끼치는 구조적 특징으로 많은 관심을 받고 있다. 또, 알지네이트 하이드로겔은 신경 네트워크에 최소한의 피해를 주면서 배양 기간 중간에 지울 수 있어 네트워크의 구조를 조절할 수 있다. 이 연구에서는 신경 네트워크의 모듈라리티를 재현하기 위해 알지네이트를 이용하여 클러스터 구조를 가진 신경 네트워크 모델을 만들고 그들의 연결의 조절 및 기능적 분석 시스템 개발을 목표로 한다. 먼저, 신경 네트워크의 기능적 분석을 위해 크로스 코릴레이션 기반의 연결도 분석법의 개발과 네트워크 버스트 모티브 검출법의 개선을 진행하였다. 또한, 이들을 적용할 수 있는 자동 분석 시스템을 구현하였다. 이후, 알지네이트 하이드로겔 패터닝을 이용해서 클러스터드 네트워크를 미세전극칩 상에 배양하고, 이들의 기능적 특성을 분석하였다. 이들은 구조가 없는 신경 네트워크와 연결도와 모티브 특성의 차이를 보이고 있었다. 화학적 패터닝과 클러스터드 네트워크의 조합을 통해 네트워크 연결을 조절하였다. 선형과 모듈형 패턴을 통해 만들어진 클러스터드 네트워크는 그 패턴대로 구조적 연결을 이루었을 뿐 아니라 기능적 연결도 만들었다는 것을 알 수 있었다.
