Capillary-assisted particle assembly (CAPA) is an attractive patterning technique that uses capillary forces for controlled and selective assembly of various nano- and microscale colloidal particles into predefined topographic templates, aimed to harness the unique optoelectronic, plasmonic, magnetic, and catalytic properties of colloids for their use in functional devices. However, CAPA suffers from poor applicability due to limited configurations of the final patterns. In this dissertation, the applicability of CAPA is expanded by diversifying the final structural configurations. First, we propose template dissolution interfacial patterning to assemble and print arrays of single gold nanoparticles onto various conductive and dielectric surfaces without the use of adhesion layers, for direct particle-substrate interactions. With direct access to the interface between the particles and substrates at hand, new possibilities arise such as refractive index-based optical property tuning, direct charge transfer for electrochemistry, and nanogap-based electromagnetic field enhancement for surface-enhanced Raman spectroscopy. Shifting gears towards the assembly merit provided by CAPA in which a wide variety of colloids can be patterned in a non-destructive manner, its applicability is expanded to biological entities that exist in the form of colloidal suspensions, namely bacterial cells. Here, CAPA is utilized to selectively pattern pathogenic bacteria at the single cell level for their identification using Raman spectroscopy. The detection platform comprises a reflective silver-silica film deposited on the topographic template to provide highly reproducible single cell signals for Escherichia coli, Staphylococcus aureus, and Salmonella gallinarum. The critically important role of CAPA is highlighted by its ability to align the most Raman active part of the cell and the potential to separate single bacteria from biologically relevant media. It is envisioned that the proposed strategies in this dissertation can open new opportunities toward the practical applications of CAPA as a single colloid patterning tool.
모세관 입자 조립은 다양한 나노 및 마이크로 규모의 콜로이드 입자를 결정론적으로 기판에 조립하는 의미있는 기술로 부상했지만, 최종 패턴의 제한된 구성으로 인해 실질적인 응용성이 떨어진다. 본 연구는 모세관 조립의 실용성을 확장하기 위해 최종 패턴 구성을 다양화하는 방법을 제안한다. 첫째, 입자-기판 상호 작용을 위해 접착층 없이 다양한 표면에 단일 금 나노 입자를 조립하고 전사하기 위한 템플릿 용해 계면 패터닝 방법으로 입자와 기판 계면에 접근함으로써, 굴절률 기반 광학 특성 조정, 직접 전하 이동을 이용한 전기 화학 분석, 나노갭 기반 전자기장 향상을 이용한 센싱과 같은 새로운 가능성을 보았다. 두 번째, 다양한 콜로이드를 비파괴적으로 패터닝하는 모세관 조립의 장점을 살려 단일 세포 수준에서 병원성 박테리아를 선별적으로 패턴화하고 라만 분광법을 이용해 식별한다. 검출 플랫폼은 은-실리카 필름이 증착된 지형 템플릿으로 구성되며 대장균, 포도상구균 및 살모넬라에 대해 재현성 높은 단일 세포 신호를 제공한다. 모세관 조립의 중요 역할은 세포의 라만 활성이 가장 높은 부분을 정렬하는 것과 생물학적 미디어에서 단일 박테리아를 선택적으로 분리하는 것에 있다. 본 연구에서 제안된 전략들을 통해 모세관 조립의 실제 적용을 위한 새로운 기회를 열 수 있을 것으로 예상된다.
