In recent decades, climate change has been striking the globe and renewable energy sources have become necessary. Biomass has been spotlighted to solve global warming and microalgal biomass especially is gaining attention with advantages regarding food-ethics and deforestation. In this study, a process cultivating a microalga species, namely Ettlia sp. YC001, was statistically optimized. The optimization increased biomass and lipid productivities by 4.9-fold and 2.4-fold, respectively. Subsequently, two feedstocks, i.e., Helianthus tuberosus and algal residue were utilized as alternative nutrient sources. They were hydrolyzed to generate low-cost medium which resulted in biomass and lipid with the same productivity. Furthermore, a sequential hydrolysis process was developed to utilize both feedstocks efficiently. Additionally, an analogous techno-economic analysis for both processes using the optimized medium and low-cost medium was conducted to examine the effectiveness of the developed processes. Hydrolysate medium decreased two-thirds and three-fourths of required operational expenditures to produce Ettlia sp. biomass and lipid, respectively. Lastly, the fructose utilization pathway of Ettlia sp. was analyzed with RNA expression levels of genes related to sugar transportation and the glycolysis pathway. The results showed that the glycolysis pathway of the species is up-regulated under the presence of fructose, and STF11, a hypothetical fructose transporter, transported fructose into cells.
최근 지구온난화가 가속화되면서 재생 가능 에너지의 필요성이 대두되고 있다. 이를 위하여 여러 세대의 바이오매스가 연구되었고, 그중 미세조류 유래 바이오매스는 식량자원이 아님과 동시에 산림파괴를 하지 않아 최근 가장 주목받고 있다. 하지만 미세조류 역시 아직 세계 에너지 수요를 맞추기 위한 생산량 및 생산단가가 미흡한 실정이다. 이를 위하여 본 연구에서는 미세조류 중 Ettlia sp. YC001의 종속 영양적 배양공정을 통계적으로 최적화하여 생산성을 극대화하였다. 그 결과 기존의 연구보다 최대 4.9 배의 바이오매스 및 2.4 배의 지질 생산성 향상을 이루었다. 그뿐만 아니라, 높아진 생산성을 유지하면서 동시에 낮은 가격으로 동질 이상의 바이오매스 및 지질을 생산할 수 있도록 Helianthus tuberosus와 조류 잔여물을 가수분해하였고 이를 위한 순차적 가수분해 공정을 개발하였다. 또한, 본 연구에서 실행한 배양 공정의 경제적 실효성을 살펴보기 위하여, 최적 생장 공정과 저가 기질을 이용한 생산공정의 경제성을 분석 및 비교하였다. 그 결과, 저가 기질을 이용한 연구는 기존 최적 배지 대비 바이오매스 생산 시 3 배, 지질 생산 시 4 배의 공정 운영 비용을 절감할 수 있었다. 마지막으로, 본 연구에서 주목한 Ettlia sp.의 과당 선호 기작을 알아내기 위하여 당 수송체 및 해당 과정 대사경로 유전자의 RNA 전사 수준을 확인한 결과, 해당 종이 과당 섭취 시 해당 과정의 대사경로가 전반적으로 상향 조절되며 STF11 당 수송체를 이용하여 과당을 섭취한다는 것을 밝혔다.
