Combination therapy with immunologic adjuvants presents a promising method to improve the efficacy of anti-OX40 cancer immunotherapy. Previous studies have shown that adjuvant-induced inflammatory signals and T cell OX40 expression can enhance OX40 stimulation by anti-OX40 mAb. However, these studies have not taken into consideration the timecourse of OX40 and expression and activity, which is a critical factor in the strength of OX40 signaling. As T cells upregulate OX40 on their surface with delayed kinetics upon activation, a delay should exist between the delivery of the activating adjuvant and anti-OX40 mAb as well. This study proposes a hydrogel-based release system for the co-delivery of STING agonists and anti-OX40 mAb. The pore size of the hydrogel system is optimized to take advantage of the size difference of the two payloads, producing a burst release of STING agonists and a delayed slow release of mAb. Local injection of this system at the tumor site will allow for anti-OX40 delivery when intratumoral OX40 expression is at its peak, maximizing therapeutic efficiency of combination therapy.
면역 보조제와의 병용 치료요법은 항-OX40 암 면역 치료의 효능을 개선하는 유망한 방법이다.면역 보조제가 일으키는 염증 신호와 T 세포의 OX40 발현이 항-OX40 항체에 의한 OX40 신호를 향상시킬 수 있음이 보고된 바 있다. 그러나 이 병용 치료요법의 기존 연구들은 OX40 신호의 강도를 결정하는 중요한 요소인 OX40의 시간별 발현 및 활성을 고려하지 않았다. T 세포의 활성화와 OX40의 발현 사이에는 시간차가 존재하기 때문에, 활성화 보조제와 항-OX40 항체의 전달 사이에도 지연이 존재해야한다. 본 연구는 STING 작용제 및 항-OX40 항체의 공동 전달을 위한 하이드로젤 기반 방출 시스템을 제안한다. 하이드로젤 시스템은 두 페이로드의 크기 차이를 이용해 STING 작용제의 버스트 방출과 항체의 지연된 방출을 유도한다. 종양 부위에 이 시스템을 국부적으로 주입하면 종양 내 OX40 발현이 최고점일 때 항-OX40 전달이 가능하여 병용 요법의 치료 효율을 극대화 할 수 있다.
