Modulation of the immunosuppressive tumor microenvironment (TME) has become the key for successful cancer immunotherapy because the suppression interferes with an effective anti-tumor immunity. Among several means of TME modulation, introducing pro-inflammatory macrophages has gained attention for their role in secreting pro-inflammatory cytokines and chemokines. Despite such anti-tumoral characteristics, macrophage-mediated modulation has been challenged by the lack of trafficking ability and immunosuppressive TME that forces anti-inflammatory repolarization. This study proposes a system of monocytes loaded with pro-inflammatory drug-encapsulating particles that modulates the immunosuppressive TME into the immunostimulatory environment. This system, benefiting from both active targeting of monocytes and continuous robustness against the pro-tumoral environment by optimized particles, is expected to create the immunostimulatory environment for effective cancer immunotherapy.
면역 억제적 종양 미세 환경이 효과적인 항암 면역 치료를 방해하기 때문에 이러한 환경을 완화시키는 것이 성공적인 치료 효과의 핵심이다. 종양 미세 환경 완화를 위한 여러 방법들 중, 염증성 사이토카인 및 케모카인을 분비하고 항원 제시 세포로 작용할 수 있다는 점 때문에 염증성 대식세포를 활용하는 연구들이 주목받아왔다. 하지만 이러한 항암 면역 특성에도 불구하고, 대식세포는 종양으로 이동하는 능력이 부족하고 면역 억제적 환경에 의해 지속적으로 항염증적 분화가 이루어진다는 한계점이 있다. 이 연구는 면역 억제적 종양 미세 환경을 면역 자극 환경으로 조절하는 염증성 약물 입자 탑재 단핵구 시스템을 제안한다. 종양 환경으로 이주하는 경향을 갖고 염증성 대식세포의 전구체인 단핵구와 염증성 분화관점에서 면역 억제적 환경에 대한 지속적인 강인함을 위한 최적화된 입자를 활용하여 효과적인 항암 면역 치료가 가능한 면역 자극 환경 조성을 기대할 수 있다.
