Dopamine is a major neurotransmitter closely related to neurological and psychological disorders. Real-time dopamine monitoring devices with external stimulation are critical to discovering the unveiled role of dopamine transport and the relationship between dopamine and the progress of neural diseases. Fast cyclic voltammetry (FSCV) has been widely used for in vivo real-time neurotransmitter detection due to its high temporal resolution. However, conventional FSCV devices, 7 μm-diameter carbon fiber microelectrode (CFME), lack multifunctionality. Even in the case of a device with multifunctionality added beyond the existing FSCV device, it showed a limitation in that it could not implement long-term usability. Here, we aim to address the discrepancy between the need for a multifunctional neural device for dopamine detection and the challenges of conventional device form using current material and fabrication methods. We developed two types of multifunctional fibers for chronic in vivo neurotransmitter monitoring: (i) carbon fiber embedded multifunctional probe (CFMP) with 200 μm-diameter, and (ii) carbon nanotube sheet wrapped multifunctional probe (CNTSMP) with 280 μmdiameter. The core optical guide part, manufactured via the thermal drawing process (TDP), consisted of polycarbonate (PC)/Poly(methyl methacrylate) (PMMA) transferring optogenetic stimulation. A flexible, multifunctional neural probe for dopamine monitoring based on FSCV was able to monitor dopamine release evoked by optical stimulation in the ventral tegmental area (VTA) in a rodent model. These flexible multifunctional probes are expected to minimize immune response to conduct a long-term multimodal study with neurotransmitter sensing.
도파민은 신경학적, 심리적 장애와 밀접한 관련이 있는 주요 신경 전달 물질이다. 외부 자극이 있는 실시간 도파민 모니터링 장치는 도파민 수송의 새로운 역할을 발견하고 도파민과 신경 질환 의 진행 사이의 관계를 발견하는 데 중요하다. 고속 순환 전압 측정(FSCV)은 높은 시간 분해능으 로 인해 생체 내 실시간 신경 전달 물질 검출에 널리 사용되어 왔다. 그러나, 종래의 FSCV 소자 인 7 μm 직경의 탄소섬유 마이크로 전극(CFME)은 다기능성이 결여되어 있다. 기존 FSCV 기기를 넘어 다기능이 추가된 기기의 경우에도 장기적인 사용성을 구현할 수 없다는 한계를 보였다. 여 기서, 우리는 도파민 검출을 위한 다기능 신경 장치의 필요성과 현재 재료 및 제작 방법을 사용 하는 기존 장치 형태의 과제 사이의 불일치를 해결하는 것을 목표로 한다. 우리는 만성 생체 내 신경 전달 물질 모니터링을 위한 두 가지 다기능 섬유를 개발했다: (i) 직경 200μm의 탄소 섬유 내장 다기능 프로브 (CFMP) (ii) 직경 280μm의 탄소나노튜브 시트 기반 다기능 프로브 (CNTSMP). 열인발 공정(TDP)을 통해 제조된 광섬유는 유연성 폴리머인 폴리카보네이트(PC)/Poly(메틸메타크 릴레이트)(PMMA)로 구성되며 광유전 자극을 전송하였다. FSCV에 기반한 도파민 모니터링을 위한 유연하고 다기능적인 신경 프로브는 설치류 모델에서 복측 분절 영역(VTA)의 광학 자극에 의해 유발되는 도파민 방출을 모니터링할 수 있었다. 이러한 유연한 다기능 프로브는 면역 반응을 최 소화하여 신경전달물질 감지를 통한 장기적인 다기능 연구를 수행할 수 있을 것으로 기대된다.
