Intrinsic fluorophores in biological sample provides structural and biochemical information within tissue. Fluorescence lifetime imaging microscopy (FLIM), which measures the fluorescence intensity and fluorescence lifetime of these endogenous fluorophores with high-resolution images, is becoming increasingly important in the biological science. However, Photobleaching occurring in fluorescence imaging and a narrow field of view of FLIM due to low image acquisition speed has limited versatility of FLIM. This study describes the development of multispectral fluorescence lifetime imaging microscopy using 1MHz pulsed laser and analog mean delay method and proposes a method for reducing photobleaching through utilization of optical property and scanning method that increase light detection efficiency. This study deals with theoretical explanation of the proposed method, the detailed design process of the system and the verification of performance through various sample experiments.
생체 내에 내재된 자가 형광은 조직 내의 구조적 정보와 생화학적 정보를 제공한다. 이러한 자가 형광 신호의 형광 세기와 형광 수명을 고해상도의 영상으로 측정하는 형광 수명 영상 현미경은 생명과학과 의학 분야에서 중요성이 점차 높아지고 있다. 하지만 형광 신호 측정에서 발생하는 광표백과 형광 수명 영상 현미경의 낮은 영상 획득 속도에 의한 작은 영상 영역은 형광 수명 영상 현미경의 범용성을 제한하였다. 본 연구에서는 1MHz 반복률의 레이저와 Analog mean delay 방식을 이용한 다채널 형광 영상 수명 현미경을 구현하고 광효율을 높이는 광학적 요소의 활용과 스캔 방식을 통해 대면적 조직의 형광 수명 측정 시 발생하는 광표백을 저감하는 방법을 제안한다. 제안한 방법에 대한 이론적 설명과 현미경의 상세 설계 과정 그리고 다양한 샘플 실험을 통한 성능 검증 등의 내용을 다루고자 한다.
