Despite having more direct information than functional magnetic resonance imaging, little is known about how the brain’s neurochemical mechanisms influence mental health and decision-making. This study, with 1H-functional MRS scanning during reward learning and computational modeling, examined whether baseline glutamate/glutamine (Glx) and lactate concentrations were related to general anxiety and depression and how they changed according to gains and losses. The glutamate concentration in the anterior insular cortex (AIC), not the medial prefrontal cortex, was positively associated with the general psychopathological factor of anxiety and depression through the mediation of error sensitivity. The AIC Glx decreased mainly after learning from gains and as the entire task was performed. Conversely, lactate reduction in AIC occurred only during learning from loss and was associated with higher general psychopathology scores. It is the first time to demonstrated that both glutamate-related error sensitivity and loss-specific lactate reduction in AIC are associated with general psychopathology. This study’s results suggest that AIC is a potential target brain region for glutamate and/or lactate-mediated therapeutics for anxiety and depression.
기능적 자기공명영상보다 더 직접적인 정보가 알려줌에도 불구하고 뇌의 신경화학적 메커니즘이 정신 건강과 의사 결정에 어떻게 영향을 미치는지에 대해서는 알려진 바가 거의 없습니다. 본 연구에서는 보상 학습 및 계산 모델링 중 수소 기능 자기공명분광법 기술을 사용하여 기준 글루탐산 및 젖산 농도가 일반적인 불안 및 우울 증상과 관련이 있는지 여부와 이득 및 손실에 따라 어떻게 변화하는지 조사했습니다. 내측 전전두엽 피질이 아닌 전섬엽 피질의 글루탐산 농도는 오류민감도의 매개를 통해 불안과 우울의 범정신병리학적 요인과 양의 상관관계가 있었습니다. 전섬엽 글루탐산의 감소는 주로 이득을 학습한 후에 발생했지만 전체 작업이 수행될 때 발생했습니다. 전섬엽의 젖산 감소는 손실로부터 학습하는 동안에만 발생했으며 더 높은 범정신병리학 점수와 관련이 있었습니다. 저는 처음으로 전섬엽의 글루탐산 관련 오류민감도와 손실에 따른 젖산 감소가 범정신병리와 관련이 있음을 보여줍니다. 이 연구는 전섬엽이 불안과 우울증에 대한 글루탐산 및/또는 젖산 매개 치료제의 잠재적 표적 뇌 영역임을 시사합니다.
