Transcription factors (TFs) move between the cytoplasm and the nucleus within a cell and regulate gene expression inside the nucleus. Homeoproteins (HPs), TFs containing the homeodomain (HD), bind to target deoxyribonucleic acid (DNA) sequences using the HD and regulate gene expression temporal-spatially to determine the cell fate during embryonic development. This inherent role as a transcriptional regulator led us to believe that HPs function in the nucleus of the cells expressing them. However, unlike general TFs, HPs have the property of migrating to adjacent cells, allowing cells to perform appropriate functions during postnatal development. In this dissertation, I give a methodology for the intracellular trafficking pathway of orthodenticle homeobox 2 (OTX2) involved in postnatal visual system maturation. First, I tracked the cytoplasmic vesicles observed near the nucleus during the nuclear egress of OTX2 and found that the lysosomes are related to the intracellular transport of OTX2. Next, based on ultrastructural analysis, I revealed that the factors involved in the vesicle formation during the nuclear egress of OTX2 are the torsin family 1 member A (TOR1A) adenosine triphosphatase associated with diverse cellular activities (AAA+ ATPase) and the linker of nucleoskeleton and cytoskeleton (LINC) complex. Finally, using mutations in these factors, I blocked the nuclear egress process of OTX2 by inhibiting the formation of perinuclear vesicles. Consequently, I found the immaturity of the visual system and the formation of OTX2 aggregates in the nucleus. Taken together, I suggest that the nuclear egress of OTX2 not only enables the proper functioning of recipient cells but also prevents aggregates-induced cell death following OTX2 accumulation within the nucleus of donor cells.
전사인자는 단일 세포 내에서 세포질과 세포핵 사이를 이동하면서 필요에 따라 세포핵 내부에 있는 유전자의 발현을 조절한다. 그중 호메오도메인을 가지는 전사인자는 개체의 발생 과정에서 형태를 결정하는 유전자의 발현을 시공간적으로 조절하고 있다. 특히, 호메오도메인 전사인자는 일반적인 전사인자와는 달리 인접한 세포로 이동하는 성질을 가지고 있으며, 이는 개체의 발달 과정에서 세포가 정상적인 기능을 수행할 수 있도록 한다. 본 학위논문에서는 호메오도메인 전사인자 중 생후 시각계 발달 과정에 관여한다고 알려진 orthodenticle homeobox 2 (OTX2) 단백질의 세포핵에서부터 출발하는 세포 내 이동 경로에 대한 방법론을 제시하고자 한다. 먼저, OTX2 단백질의 세포핵 분비 과정에서 관찰되는 소포를 추적하여 세포 내 소기관 중의 하나인 리소좀이 OTX2의 세포 내 이동과 연관되어 있음을 확인하였다. 다음으로, 세포핵 소포의 초미세구조 분석을 바탕으로 소포 형성에 관여하는 인자가 다양한 세포 활동과 연관된 아데노신 삼인산 가수 분해 효소 (adenosine triphosphatase associated with diverse cellular activities; AAA+ ATPase)의 하나인 torsin family 1 member A (TOR1A), 그리고 세포핵과 세포골격을 연결하고 있는 linker of nucleoskeleton and cytoskeleton (LINC) 복합체임을 규명하였다. 마지막으로, 이들 인자의 돌연변이형을 이용하여 세포핵 소포로 매개되는 OTX2의 분비 과정이 정상적으로 이루어지지 않는 환경을 만들었고, 결국 시각계의 미성숙 현상과 OTX2 단백질의 세포핵 내부 응집체 형성을 확인할 수 있었다. 이러한 결과는 OTX2 단백질의 세포핵 분비 현상이 공여 세포의 정상적인 기능을 가능하게 하는 것뿐만 아니라, 수여 세포 내에 존재하는 OTX2 단백질의 수용 범위 이상의 축적을 방지함으로써 응집체 형성으로 기인하는 세포 사멸의 회피를 위한 과정의 하나라는 점을 시사한다.
