Despite of its importance and severity, the pathophysiology of neuropsychiatric disorders remains in hidden. Recent advances of analytic technologies lead mass production of high-quality multi-omics data and data-driven research. In this dissertation, we developed data-driven methods to explain the pathophysiology by combining data analysis including deep learning and suggested new aspects of neuropsychiatric disorders.
First, we developed novel network comparison method for gene coexpression network. We applied new method to comparative analysis between Huntington’s disease (HD) and brain aging and found that HD and aging have common aspects in gene expression related to neurodegeneration and immune response. Second, we developed step-wise deep learning with multi-precision data technique and constructed an interpretable network model between genotypes and schizophrenia. We found novel combinatorial SNP marker of schizophrenia and proposed relationship between neuronal growth and schizophrenia susceptibility by analyzing the model. I expect that data-driven approaches to neuropsychiatric disorders like this dissertation will expand our understanding on the diseases.
신경정신질환은 중요성과 심각성에도 불구하고 그 병태생리가 미지에 있다. 최근 분석기술의 발전은 고품질 멀티오믹스 데이터의 생산과 이를 활용한 데이터 기반 연구로 이어졌다. 본 연구는 멀티오믹스 데이터에 심층학습을 비롯한 데이터 분석을 조합하여 병태생리를 밝히기 위한 방법론들을 개발하고 이를 신경정신질환에 적용하여 질병의 새로운 측면들을 제안하였다. 유전자 공발현 네트워크를 비교하기 위한 비교방법론을 개발하고, 이를 헌팅턴병과 뇌의 노화를 비교하는데 적용하였고, 헌팅턴 병과 노화가 퇴행성과 면역과 관련된 유전자 발현을 공유하고 있음을 규명하였다. 다음으로 다수준의 데이터를 활용한 순차적 심층학습 방법론을 개발하여 유전형과 조현병간의 해석가능한 네트워크 모델을 제안하였다. 모델을 분석하여 조현병의 새로운 마커를 찾고, 조현병의 선천적 요소가 신경세포와 발달과 관련이 있음을 제안하였다. 이와 같은 신경정신질환에 대한 데이터 기반의 접근이 질환에 대한 이해의 폭을 넓힐 것으로 기대한다.
